| Purpose:With the help of adriamycin nephropathy model, we investigated the intensity effect of Fang ji Huang qi Decoction on renal injury of rats with adriamycin nephropathy model, and discussed the pharmacodynamic mechanism of Fang ji Huang qi Decoction on adriamycin nephropathyMaterial and method:72SD rats were randomly divided into6groups according to weight, the blank group, model group, Chinese medicine control group, Fang ji Huang qi Tang high.middle and low dose group. Except the blank group, the rest rats were all given adriamycin in the dose of5mg/kg body weight dose once a day for3days by tail vein injection to establish the adriamycin nephropathy model. And then, all of the rats were filled Fang ji Huang qi Decoction rather than the blank group once a day for24days. The amount of24h urinary protein were detected on the days of Od,7d and21d of the treatment.At the end of the experiment, the content of BUN, Alb, SCr TP, TG, TC were tested by automatic biochemical analyzer, weight method was used to test KWI, and ELISA were applied to detect IL-6.TGF-β1,CTGF concentration of rats renal tissue.Results:1. The effect of Fang ji Huang qi Tang on SCr, BUN, quantitative of24h urine protein, KWI and urine volumeCompared with the normal group, Cr, BUN.24h urine protein quantitative and KWI of model group rats were significantly increased, but urine volume significantly decreased (P<0.05).Compared with the model group, Fang ji Huang qi Tang Gao. in low dose group, SCr, BUN.24h quantity of urinary protein was significantly reduced, KWI decreased, urine volume increased significantly (P<0.05).2. The effect of Fang ji Huang qi Tang on Alb, TP, TG, TCCompared with that of the normal group, Alb, TP, TG and TC of model group were all obviously increased (P<0.05).Compared with the model group, Alb of low dose group of Fang ji Huang qi Tang. TP of medium and low dose of Fang ji Huang qi Tang. TG of high, medium and low dose of Fang ji Huang qi Tang Gao, TC of high and low dose of Fang ji Huang qi Tang were all decreased obviously (P<0.05). 3. The effect of Fang ji Huang qi Decoction on IL-6, TGF-β1, CTGF concentrations of rats renal tissueCompared with the normal group, IL-6, TGF-β1,CTGF concentration of rats renal tissue in model group remarkably increased (P<0.05).Compared with model group, IL-6concentration of high, medium and low doses of Fang ji huang qi Tang, low-dose renal tissue decreased, traditional Chinese medicine in the control group, CTGF concentration of Chinese medicine control group, high, medium and low doses of Fang ji huang qi Tang remarkably decreased (P <0.05).Conclusion:1. Fang ji Huang qi Tang can reduce SCr, BUN and KWI of rats with adriamycin-induced nephropathy, decrease the quantity of24h urinary protein, increase urine volume of rat with nephropathy and has a certain protective effect on renal function of rats with adriamycin nephropathy.2. Fang ji Huang qi Tang can enhance serum Alb and TP of rats with adriamycin nephropathy, decrease TG and TC, and has an auxiliary role in improving renal function.3. Fang ji Huang qi Tang plays the role in reducing kidney damage and protecting kidney by reducing IL-6and CTGF concentrations of renal tissue, improving IL-6and CTGF concentration of rats with adriamycin nephropathy. |
