Semen Ziziphi Spinosae, botanically from the dried seeds of Ziziphus jujuba Mill.var. spinosa (Bunge) Hues. H.F. Chou is a traditional tranquilizing medicine frequently used in China for the treatment of a variety of syndromes and diseases, including insomnia, neurasthenia.The paper is to study on the chemical constituents and content determination of main saponins which includes five chapters.Chapter one:summary of Semen Ziziphi SpinosaeOrigin, distribution, chemical constituents, pharmacological activities and quality control method of Semen Ziziphi Spinosae was summarized. For this article, it provided some reference of topic basis, determining the research content and Innovation points.Chapter two:studies on the chemical constituents of Semen Ziziphi Spinosae.The ethanol crude extract of Semen Ziziphi Spinosae was separated by silica gel column chromatography.The prefractionation was further separated by ODS, medium pressure liquid preparation and purified by Sephadex LH-20. Nine compounds among the eleven isolated constituents of Semen Ziziphi Spinosae were identified by UV, MS, NMR as follows: spinosin,6"’-feruloylisoSpinosin, jujuboside A, jujuboside Al, jujuboside B, jujuboside B1, zizyphus saponin Ⅲ, daucosterol, ferulic acid. Among the compounds, zizyphus saponin Ⅲ was isolated from Semen Ziziphi Spinosae for the first time.Chapter three:studies on the quality of main saponins.Inspected purity, stability of jujuboside A, B. TLC and HPLC methods were used for purity inspected of jujuboside A, B. TLC showes the two substance has no any impurity spot and HPLC indicates that it’s purity reaches98.0%above.The results of stability show that the two substance are relatively stable under the conditions of temperature(60℃),light(4500±500Lx) and H2O2.Chapter four:Simultaneous determination of jujuboside A, jujuboside B, jujuboside Al and Bl in semen Ziziphi spinosae by high performance liquid chromatography(HPLC).Jujuboside A, Jujuboside B, Jujuboside A1and Jujuboside B1in semen Ziziphi spinosae were determined by HPLC. Jujuboside A had a favourable linear relationship in the range of0.2-6.6μg and the regression equation was as follows:Y=283095x-5283.8, R2=0.9997(n=6),the contents of jujuboside A in different origin of semen Ziziphi spinosae were Shandong(0.10%), Hebeizanhuang(red)(0.081%), Heibeizanhuang (black)(0.076%), hebeianguo(0.046%),Shanxi(fried)(0.042%),Shanxi(crude)(0.042%),Hunan(0.028%),Hebeilo nghua(0.019%),Yunan(0.0076%),Miandian(0.0043%) andYuenan((0.0042%)); Jujuboside B had a favourable linear relationship in the range of0.5-4.2μg and the regression equation was as follows:Y=343700x-4712.4, R2=0.9995(n=6),the contents of jujuboside B in different origin of semen Ziziphi spinosae were Hebeizanhuang(black)(0.066%), Heibeizanhuang(red)(0.051%),Shandong(0.041%),Shanxi(fried)(0.030%),Shanxi(crude)(0.029%),hebeianguo(0.026%), Hunan(0.021%), Yunan(0.012%), Yuenan(0.012%),Miandian(0.011%),Hebeilonghua(0.011%); Jujuboside A1had a favourable linear relationship in the range of0.06~0.6μg and the regression equation was as follows: Y=292143x+6647.3,R2=0.9994(n=6),the contents of jujuboside A1in different origin of semen Ziziphi spinosae were Shandong (0.0094%),Hebeizanhuang (black)(0.0088%), hebeianguo(0.0073%),Heibeizanhuang(red)(0.0061%),Shanxi(fried)(0.0060%),Shanxi(crude)(0.0059%),Hunan(0.0055%),Miandian(0.0037%),Hebeilonghua(0.0032%),Yunan(0.0024%),Y uenan(0.0013%); Jujuboside B1had a favourable linear relationship in the range of0.036~0.48μg and the regression equation was as follows:Y=380672x+6109.6,R2=0.9993(n:=6),the contents of Jujuboside B1in different origin of semen Ziziphi spinosae were Hebeizanhuang(black)(0.0055%),Hebeianguo(0.0041%), Shandong(0.0035%),Heibeizanhuang(red)(0.0030%),Shanxi(fried)(0.0020%), Shanxi(crude)(0.0019%),Hunan(0.0017%),Yuenan(0.00076%),Miandian(undetected),Hebeilo nghua(undetected),Yunan(undetected); the results showed jujuboside A, jujuboside B jujuboside A1and jujuboside B1in different origin of semen Ziziphi spinosae had differences.
