In order to search for the gene marker for the elite endurance athletes, our research studied the polymorphism sites of tryptophan hydroxylase (TPH) gene and the haplotype of TPH, we also studied the possible association between genotype of the four single nucleotide polymorphisms (SNP) or haplotype of TPH gene and cardiopulmonary physiological indexes.I:Case-Ccontrol StudyThe case-control study used PCR-RFLP to test the polymorphism of A218C A21328G, A40237G and-C8347G in order to analyze the genotype or allele distribution of the four sites and the haplotype of TPHI and TPHII between Case-Control groups. Case group included180Northern Chinese long-distance runners and control group included333controls.Results:TPHI:1) The frequency of GG genotype of A21328G was0.603in the athlete groups, it was significantly (p<0.05) higher than controls (0.486);2) The frequency of A-C haplotype was0.225in the athlete groups, it was significantly (p<0.05) lower than controls (0.302).TPHII:The frequency of C-G and G-A haplotype in the male athlete groups were significantly higher than controls, but the frequencies of the two haplotypes were the lowest in the athlete groups.II:Cross-Sectional Association StudyUsing PCR-RFLP to test the polymorphism of the above four sites in the group which included63long-distance female runners who were still in service, then analyzed the possible association between genotype of the four SNPs or haplotype of TPH and cardiopulmonary physiological indexes.Results:TPHI:1) The Vital Capacity (VC) of athletes who had the AC genotype of A218C was3.83±0.36L, it was significantly (p<0.05) higher than controls (3.52±0.25L);2) The VC of athletes who had the AG genotype of A21328G was4.01±0.45L, it was significantly (p<0.05) higher than controls (3.74±0.34L). The Maximal Oxygen Uptake/Weight (V02max/weight) of athletes who had the AG genotype was61.2±4.9mL·kg-1·min-1, it was significantly (p<0.05) higher than controls (55.4±3.1mL·kg-1·min-1);TPHII:The Hap-Score of V02max of G-A haplotype was-2.187, it was significantly (p<0.05) lower than the mean (0).Conclusion:1) TPHI gene:The GG genotype of A21328G could be considered as the gene marker for Northern Chinese elite endurance athletes; The A-C haplotype had relationship with profermance of Northern Chinese elite endurance athletes; The A218C had relationship with vital capacity of Chinese female elite long-distance runners; The A21328G had relationship with vital capacity and V02max of Chinese female elite long-distance runners;2) TPHII gene:The G-A haplotype had relationship with V02max of Chinese female elite long-distance runners.
