Objective:To explore the intervention and interaction of aerobic exercise and KGM on the form of insulin resistance induced by high-fat diet in rats, and discuss mechanism of the form of insulin resistance and the intervention by aerobic exercise and KGM.Methods:The study objects were506-week-old male SD rats, weight180-220g. One week of adaptive feeding, then the50rats were randomly divided into5groups, those were, control group (C), high-fat group (HF), high-fat with KGM group (HK), high-fat with exercise training group (HE), high-fat with exercise and KGM group (HEK). The C group were fed with ordinary diet, while the other4groups were all fed with high-fat diet. At the same time, the HF group didn’t take exercise training, the HK group added KGM, the HE group took exercise training, and the HEK group added KGM and took exercise training; Exercise training were used60minutes’ unloaded swimming for11weeks,6times per week; the adding dosage of KGM was50mg per kilogram weight,1time per day.11weeks later, drew materials on next morning of the last exercise training and keeping fasting. FPG、FINS、 APN、AdipoR2、PPARα、TC and TG in each group of rats were examined.Results:①Compared with C group, the FPG、FINS contents and HOMA-IR increased significantly (P<0.01) as well as the ISI (P<0.01) decreased significantly in HF group rats. meanwhile, the contents of serum and hepatic TC and TG increased significantly.②Through the double factor variance analysis, aerobic exercise could lower the level of FPG(P<0.05)、FINS(P<0.01)and HOMA-IR(P<0.01),increase the ISI(P<0.05)by high-fat diet in rats, in addition, it decreased the contents of serum TC (P<0.05),also hepatic TG (P<0.05)and TC(P<0.01)contents decreased remarkably, however the serum TG (P>0.05) contents had no conspicuous difference. Supplying KGM could conspicuously decreased FPG(P<0.05)、FINS(P<0.01)contents and HOMA-IR(P<0.01), increased the ISI(P<0.05), and it could decreased the serum TC (P<0.01) and TG (P<0.01) contents as well as hepatic TG (P<0.01). Although KGM combined with aerobic exercise could make FPG and serum TC concentration to be further decreased, but it had no significant difference (P>0.05).but to FINS (P<0.01), HOMA-IR (P<0.05) and ISI (P<0.05), they had remarkable interaction, and it can make the FINS and HOMA-IR decreased further. Aerobic exercise combined with KGM could decrease hepatic TG (P<0.05)、TC (P<0.01) and serum TG (P<0.01) by high fat diet in rats, which had a significant interaction.③ompared with the C group, hepatic APN、PPARa contents (P<0.01) and serum APN (P<0.05)decreased significantly in HF group, but the content of hepatic AdipoR2(P>0.05) increased in HF group. Through the double factor variance analysis, we can conclude that aerobic exercise can significantly increased the content of hepatic APN (P<0.05) and PPARa (P<0.01) as well as the content of serum APN (P<0.01), otherwise, it decrease the content of hepatic AdipoR2(P<0.01); Supplying KGM can significantly increased the contents of hepatic APN (P<0.01)、PPARa (P<0.05) and the content of serum APN (P<0.05), although the content of hepatic AdipoR2reduced,it had no statistical significance (P>0.05). Aerobic exercise combined with KGM could make the serum APN、the contents of hepatic PPARa and APN to be further increased, and it decrease hepatic AdipoR2, they had conspicuous interaction (P<0.05)Conclusion:①11weeks of high-fat diet could induce the formation of insulin resistance in rats, the disorder of lipid metabolism induced by high fat diet, which may be the important reasons for IR.②Aerobic exercise or KGM could significantly prevent the formation of insulin resistance in rat by high-fat diet, and KGM combined with aerobic exercise had significant interaction on preventing the formation of insulin resistance induced by high-fat diet in rats. The mechanism may be related with that aerobic exercise or KGM effectively improved blood lipids metabolism by high fat diet in rats.③Aerobic exercise or KGM may modulate blood lipids metabolism by hepatic Adiponectin/PPARa signaling pathways of the rats which is high fat diet, thus it could effectively prevent the formation of insulin resistance induced by high-fat diet in rats.Meanwhile, KGM combined with aerobic exercise had significant interaction to improving hepatic adiponectin/PPAR-α in high-fat diet rats.
