Gene expression is regulated by the various comprehensive factors, it is a highly complex process. In the biological process of development and growth, DNA sequence of genes is in the same condition, while the expression level is quite different. Life genetic information is not only determined by DNA, but also determined by the epigenetic bearing the same genetic material. Epigenetics is the important content in gene expression regulation mechanism, aiming at the main pattern of epigenetic regulation of histone modification, we has conducted a lot of research work. Based on the correlation between histone modifications and gene expression, this paper studied relation of the gene expression levels in human H1cell line and histone modifications.First of all, we analyses the distribution characteristics about the26kinds of histone modifications in genome-wide higher expressed genes and lower expressed genes of promoter, exon and intron areas. The results show that histone modification levels in these three functional areas of higher and lower expressed genes are different. The content of H3K27me3in promoter, exons and introns these three functional areas of higher expressed gene was obviously lower than the content of higher expressed genes. At the same time, we found that content distribution of a particular histone modifications in three functional areas of the two classes of genes is not consistent. All the acetylated modification content distribution in higher expressed genes of three functional areas, the promoter is the highest level. However, the exon is highest in the lower expressed genes.Secondly, we analyzed the distribution characteristics about11histone modifications of promoter, exons and intron area of higher expressed and lower expressed genes in chromosomes4.We divided the gene promoter into20windows, exon10windows, intron10windows. The study have found that the content about each histone modification in promoter, exons and introns these three functional areas of higher expressed genes is different from lower expressed genes. At the same time, we also found that the content distribution of a particular histone modifications about the two classes of genes is not consistent. This paper research, not only confirmed the histone modification can regulate gene expression, and the regulative effect is of regional specificity. At the same time, also confirmed the distributions about histone modifications in each functional area of the higher expressed genes and lower expressed genes is not consistent.
