Transcription factors,histone modifications,and their interactions play an important role in the regulation of gene expression.The selective binding of transcription factors is regulated by the histone modification.Transcription factors can also regulate the expression of target genes.The regulation of transcription factors is not only related to the position of binding to the gene and its function,but also related to the binding mode of the transcription factors.Different binding modes will have different effects on the regulation of the target gene.When transcription factors accurately regulate target genes,they tend to bind to other transcription factors in regulatory regions.These transcription factors can be either cooperative or competitive.In this paper,two types of mouse cell lines are selected,which are mouse leukemia cells MEL and mouse B lymphocytes CH12.The narrowpeak data of 29 transcription factors from two cell lines in the ENCODE database were selected and study on the issues of co-binding of transcription factors.Because the transcription factors with higher overlap rates be inclined to high probability ofco-binding rates,it is necessary to find the overlapping rules of transcription factors.Using statistical methods to analyze the relationship between transcription factors and histone modifications and then co-binding of transcription factors were classified and predicted using histone modifications as a feature,finally,H4K4me1,H2K9 ac and K3K36me3 were found the more reliable predictors.In order to further screen the peak data of co-binding of transcription factors,we studied the motif of co-binding of transcription factors which have specific regulatory relationships,these are c-Myc+Max and Mxi1+Max.Based on the distance between the midpoint of the binding peak and the transcription start site,the target gene was predicted and analyzed using an exponential decay BETA model.The expression of the target gene in the two cell lines was compared to obtain the differentially expressed genes of cell line-specificity.We found that the up-regulated genes in cancer cells are involved in the regulation of their own immune system relative to the normal cells.This study provides a reliable theoretical basis for the study of gene expression regulation of transcription factor co-binding,and further reveals the specific regulation of transcription factor co-binding in cancer.
