In our study, three typical kinds of wood-rotting fungus from Hat Mountain Experimental Forest Farm of Northeast Forestry University were used as specific material, using the length-measuring and the mycelia of net-weight method to compare the three phellinus in the solid culture medium at the growth rate, what is more, testing the biomass of liquid medium culture. In order to compare ligninolytic enzyme in sawdust induced by colorimetric detection, aiming at comparing cellular enzyme biological activity with different carbon resources performed by DNS method. Meanwhile, using TRAP marker was regarded as a novel method to analyze the morphology for Analysis of the genetic diversity and the difference about lignin and cellular coding genes of three kinds of wood-rotting fungi by comparation. Therefore, TRAP molecular markers could be applied to genetic analysis of wood-rotting fungi.The growth speed of wood-rotting fungi in the PDA solid medium is rather uniform, Fomes fomentarius and Phellinus igniarius have relatively fast speed, compared to Phellinus linteus. However, all of them were identified as the fast growth-type.By Comparing the growth of wood-rotting fungi in liquid medium, the biomass of Fomes fomentarius is the highest, followed by Phellinus linteus, as a result, that of Phellinus igniarius is the lowest. The adjustment of growing-cycle for Fomes fomentarius and Phellinus linteus is relatively shorter, while growth-rate in the logarithmic phase was faster. Exactly, the tendency of growing cycle and growth-rate had oppositive direction.The effect of sawdust induced by ligninolytic enzymes from three typical kinds of wood-rotting fungus is different, sawdust-inducing has a effect-promoting on Fomes fomentarius and Phellinus linteus via the expression of Lip, while, not significant discrepancy; sawdust-inducing has no effect-promoting on Phellinus igniarius. The difference on the LiP from three typical kinds of wood-rotting fungus is not significant in comparation with control group, finally, we also got the same conclusion in control group. However, the expression of MnP obtained from Fomes fomentarius%Phellinus linteus and Phellinus igniarius inducing has a effect-promoting on sawdust. The difference on the MnP from three typical kinds of wood-rotting fungus is significant in control group compared to the three mentioned samples, the expression of Lac from Fomes fomentarius、Phellinus linteus and Phellinus igniarius also got the same trendency in discrepancy. To sum up, the difference in marker gene expression took on oppositive direction for Fomes fomentarius、Phellinus linteus and Phellinus igniarius.The effect of different carbon resources induced by cellular enzyme activity is significant. the expression of EG and EC from the three typical kinds of wood-rotting fungus is significant in two kinds of culture medium; but the expression of EG and EC is not significant when regarding sawdust as carbon source, after testing the activity, we conclude that biological activity is low, but the variety of activity is subtle. The expression of EG and EC is extremely significant when regarding wheat bran as carbon source, the activity is high, the variety of activity is great.Based on the detection of amplified products, A total number of29pairs of primers were identified from strip map. Including4pairs of primers to tag CBH I encoding gene,6pairs primers to tag CBH Ⅱ encoding gene,3pairs primers to tag the CDH encoding gene,1pair primers to tag the EG encoding gene,6pair primers to tag the Lac encoding gene,8pair primers to tag the LiP encoding gene, as well as1pair primers to tag the MnP encoding gene.All in all, the results demonstrated that three kinds of wood-rotting fungi tagged by TRAP-PCR generated357bands, among of them255are the polymorphic bands, accounting for71.43%of total bands, stripe size range from100to2000bp.
