The purpose of the present work was to extract, isolate and purify the flavonoidsfrom Crataegus pinnatifida bge (hawthorn), and analyse the component of flavonoidscompounds, which will be of great significance for further utilization andpharmacological investigation of hawthorn.First, extraction of flavonoids compounds from hawthorn by microwaveextraction, supercritical extraction and accelerated solvent extraction. Single factorexperiment is done, and on the basis, the orthogonal test was carried out to getrespective optimal conditions. The optimum conditions of microwave extractionmethod is as follows:the ratio of material to liquid of1:30, the concentration ofethanol of70%, extraction time of1.5min,microwave power of320W, and theextraction rate under the optimum conditions was3.64%. The optimal conditions forthe supercritical extraction method is as follows: extraction temperature of45℃,extraction pressure of25MPa, entrainer dosage of20ml ethanol of70%every10graw material, extraction time of90min, and under it, the extraction rate of flavonoidswas3.80%. The optimal conditions for accelerated solvent extraction methodextraction is as follows: extraction temperature of80℃, extraction time of3min,circulation of3times and solvent of50%ethanol. Under the conditions, thehighest extraction rate of flavones from hawthorn was4.99%.The flavonoids compounds in crude extract from hawthorn were separated bypetroleum ether, ethyl acetate, n-butanol and purified by silica gel column. Theisolated components were analyzed by GC-MS and LC-MS.GC-MS detected20kinds of compounds, of them11kinds of compounds were first discovered inhawthorn. LC-MS detected129kinds of compounds, and there were29kinds ofcompounds first discovered. Identified12kinds of flavonoids, and5of them werefirst found in hawthorn.
