| Rotala rotundifolia is a multi-ecotype aquatic plant,which can grow as the emerging plant,emersion plant and submerged plant,and showed highly absorption ability towards nitrogen and phosphorus that giving it the potential to repair eutrophic water bodies.Moreover,it is tranditionally used as a Chinese herbal medicine,thus,it is promising to construct a favorable remediation-resource utilization cycle by the application of Rotala rotundifolia in water ecological restoration and subsequently extraction of the functional comments after harvest.As the fundamental research,the presented study qualitatively and quantitatively evaluate the active ingredients of Rotala rotundifolia for the first time,and the variations of ingredients amounts in root,stem,leaf and whole plant with growth time in different eco-types(emerging and submerged states)were investigated.Then the total flaconoids was selected as the major object,and the optimal extraction process,separation and purification process parameters were evualted.The main conclusions are as follows:(1)The active ingredients in Rotala rotundifolia were identified as total flavonoids,tannins,total triterpenes,total alkaloids.In terms of emerging type,the contents dry weight of total flavonoids,tannins,triterpenoids and alkaloids in aerial parts were 126.36 mg·g-1,16.43 mg·g-1,66.65 mg·g-1,13.92 mg·g-1 respectively,which were 15.68 mg·g-1,7.68 mg·g-1,23.31 mg·g-1,2.17 mg·g-1,respectively in the underground parts.(2)In the case of emerging state,the optimal harvest time is the 90th day after planting for total flavonoids extraction,which becomes to 180,150 and 30 days for extraction of tannin,total triterpene and total alkaloid respectively.In terms of submerged state,the optimal harvest time is the 30th day after planting for total flavonoids and total alkaloids,which becomes to 90 days for extraction of tannin and total triterpenes.(3)Ultrasonic extraction shows higher extraction rate and the optimum parameters in extraction process of total flavonoids from Rotala rotundifolia is as follows:the extraction power is set up as 333 W,and extraction process is running for 50 min with the set up solid-liquid ratio(1:60),and ethanol volume fraction(79.3%);Under these conditions,the amount of total flavonoids in extraction products was as high as 32.33 mg·g-1.(4)The optimum conditions for the separation and purification of total flavonoids from Rotala rotundifolia were as follows:HPD 100 macroporous resin,injection flavonoids 2.5 mg/mL,loading volume 35 mL,ethanol volume 70%,elution volume 95 mL,adsorption and desorption temperature 50℃;Under the optimal technological conditions,the average purity of total flavonoids is 76.0%,and the average recovery rate is 86.4%. |
