| Saccharomyces boulardii isolated from Indonesian lychee was a probiotics, whichbelonged to a subspecies of S.cerevisiae. The pharmaceutical preparations ofSaccharomyces boulardii can effectively reduce the concentration of the pathogenicbacteria, degradate bacterial toxins, maintain the body microecological balance andstrengthen the immune system. It has been widely used to treat a variety of diarrhea inhumans and animals, and it has achieved good efficacy. The Saccharomyces boulardiipreparations was used to treat animal diarrhea, which is the inevitable trend in theanimal husbandry. It would have a broad application prospect. This paper firstlyscreened medium and optimized fermentation conditions in order to obtain highconcentrations of bacterial fermentation broth. Then we prepared three kinds ofSaccharomyces boulardii probiotics, which are freeze-dried powder, hollow alginatemicrocapsules and freeze-dried microcapsules microcapsules.The main contents and results are as follows:1. Screening of culture medium and fermentation condition optimization.we screened six commonly yeast culture medium. we found that the wort mediumhas high production and suitable for industrial production. Then we optimized thefermentation conditions. The result is:the incubation temperature32℃, the mediumpH natural, the concentration of sugar wort10Brix, liquid volume100mL/500mL,inoculum size2%.2. Preparation of lyophilized powder.In order to obtain high activity Saccharomyces boulardii powder, we optimizedthe cryoprotectants recipe by orthogonal test method. The best cryoprotectants recipe isskim milk powder (10%), trehalose (12%), glycerol (6%), sucrose (10%). The mixingconditions of yeast cells and cryoprotectants is: the pH of the protecting agent5, themixing ratio of the protecting agent and the bacterial2:1, the equilibrium time60min.Under this condition, we get the the maximum amount of live yeast cells, reaching1.96×1010cfu/g.3. Preparation of hollow calcium alginate microcapsules.In order to obtain a smaller particle size, spherical shape, higher mechanicalstrength and higher encapsulation yield of hollow alginate microcapsules, weoptimized its preparation process. The optimization result is: CaCl22%, CMC2%,60-80drip drops/min, sodium alginate1.2%(w/v), the film forming reaction time 15min. Under such conditions, the survival cells reached1.2×108cfu/g and theencapsulation capacity reached87%. The microcapsules obtained has good tolerabilityin artificial gastric juice, artificial intestinal fluid, artificial bile, high osmoticenvironment, but is not stable during storage.4. Preparation of lyophilized microcapsules.To get more easily storage and transport Saccharomyces boulardii microcapsules,we prepared lyophilized Saccharomyces boulardii microcapsules. We optimized theprocess in the capacity of encapsulation as an indicator. The results are: sodiumalginate concentration2%, CaCl21%, emulsifying time10min, the curing time15min.Under such conditions, the capacity of encapsulation reached67%and the survivalcells reached7.9×107cfu/g. It has good tolerability in artificial gastric juice, artificialintestinal fluid, artificial bile, high osmotic environment. Moreover It is stable duringstorage and transport. |
