Houttuynia cordata Thunb is a kind of edible and medical plant, which has antibacterial and antiviral effect. However, the scale of the comprehensive utilization of Houttuynia cordata lead to lager excavation, destroyed resources, fragmented habitat and shrinking gene pool. Then, new cultivar breeding and scale cultivation had become a great demand of Houttuynia cordata industry development.The rapid propagation technology of varieties of seedlings,which was the basis of Houttuynia cordata scale cultivation.But the small propagation coefficient of the traditional cutting and ramets propagation method,was difficult to meet the production requirement, and the research of tissue culture research on Houttuynia cordata. was little. The purpose of this study was to establish the rapid propagation system of Houttuynia cordata Thunb,which based on the tissue culture,and evaluate the genetic stability for regenerated plants.The results were as follows:1. Established the rapid propagation system of Houttuynia cordata Thunb(1) The best sterilization approach was to sterilize explants using streptomycin and penicillin (0.5mg/L) with12hours and alcohol(70%) for30s firstly, then using HgCl2(0.1%) for12minutes, after washed4to5times by water, and HgCl2(0.1%) for10minutes respectively.(2) The best condition for the germination and growth of adventitious buds was adding0.5mg/L NAA and2.0mg/L6-BA to MS medium,induction rate could be as high as96.65%.(3)By the single factor test, the best mediun was adding4-5mg/L GA3to MS, and the proliferation rate was as high as560~563%; The orthogonal experiment result showed that MS+1.5mg/L6-BA+5mg/L GA3+0.2mg/L NAA was best and the proliferation rate was917%, while the effect from large to small in turn was6-BA, GA3and NAA on proliferation.(4) MS+1.0mg/L NAA had the best promotional effect on improve rooting.(5) The optimal transplanting medium was cultivated soil and vermiculite(1:1), which could result in99%survival rate and healthy grown regeneration seedlings. 2. Evaluation of the genetic stabilityWith the common propagated plantlets (Common plantlets) as contrast, genetic stability of Tissue culture plantlets were studied.①Through the observation of agronomic character, common plantlets and tissue culture plantlets were with the same morphology;②At the cellular level, there was no difference between tissue culture plantlets and common plantlets on cell ploidy;③At the molecular level, the SRAP markers results showed that Houttuynia has some slight difference within tissue culture plantlets, but it was infrequent and unobvious. Tissure culture plantlets could be stability for genetics.
