| Houttuynia cordata Thunb is perennial herbs of Houttuynia Thunb of Saururaceae,the whole plant belong to the plant resources"medicine and food dual-purpose".With the comprehensive scale utilization to it,it was excessive excavation;the wild production has been in short supply.This experiment used respectively the stems and rhizomes of Houttuynia cordata Thunb as explants,studied the effect factors of the sterilization time,medium,hormone and so on to tissue culture,and preliminarily established an efficient regeneration system and provide a theoretical basis for the establishment of Houttuynia cordata Thunb rapid propagation system.The results of the experiment are as follows:1.Best basic medium and explant:in ME,-ME8 treatment,the highest treatment of the germination rate is the ME3,thus determine best basic medium is MS medium and the best explant is the annual underground stems of Houttuynia cordata Thunb.2.Explant sterilization:the best sterilization approach was using streptomycin and penicillin for 05 hours,then using 0.1%HgCl2 for 20 minutes.3.Adventitious buds induction:The best medium of adventitious buds induction is MS+0.5mg/LNAA+2.0 mg/L6-BA+1.0mg/L KT,induction rate reached high to 96.85%.4.Adventitious buds proliferating:?useed single NAA,6-BA,GA3 and concentration were respectively 0.50mg/L,1.Omg/L and 4-5mg/L,the proliferation rate of adventitious bud reached maximum;?used meanwhile them,MS+0.5mg/L NAA+1.5mg/L 6-BA+4mg/L GA3 is the optimal proliferation medium.5.Adventitious buds proliferating:the plantlets rooting rate?100%?and root diameter?1.34mm?of 1/2MS medium significantly higher than that of MS medium?94.33%and 0.81mm?;1/2MS+1.0 mg/L IAA is the best rooting culture medium.6.Domestication culture:the optimal transplanting medium was cultivated soil and vermiculite,and survival rate reach 99%.7.Transplanting culture:with processing in different light quality,the fresh weight of plant is blue light>red light>yellow light>white light>green light. |
