| Background:Recently, with increasing pressures of life, natural disasters and other non-combatmilitary emergency tasks, stress-related diseases are increasing. stress ulcer is the mostobvious, which seriously affect people’s quality of life and the combat effectiveness.Studies had shown that stress ulcer was the result of a combination of several factors,including the neuroendocrine regulation, the relative enhancement of mucosal injuryfactors (such as gastric acid, pepsin, bile), the weakening of the role of the gastricmucosal defensive barrier and so on. Epithelial cells and tight junctions was the most important factors in the gastric mucosal barrier. In recent years, it had been found thatadhesion junction played an important role in the connection of the cells andextracellular matrix, which regulated epithelial cell migration, proliferation andapoptosis, through a complex signaling pathway. The study of relation betweenadhesion junction and stress ulcer is still blank, and the role of adhesion junction in therepair of gastric mucosal injury is also unknown. Hemostatic gel, of which mainingredients are Baiji and Ejiao has a significant effect on gastrointestinal bleeding,because it not only has haemostatic effect, but also can adhere to the mucosa of erosionand bleeding. so it has important practical significance to study the effect of adhesionjunction in the pathogenesis of stress ulcer, and it will provide a theoretical evidencefor prevention of stress ulcer.Objective:Firstly, by using the sleep deprivation method, animal models of stress ulcer inrats were set up. With observed the general samples of gastric and detected the serumlevel of EGF, IL-2, PAF, it was confirmed that the small platform over water methodcan successfully build stress ulcer model. Secondly, to analysis of the correlationbetween VCL, FAK and gastric mucosal injury, the expression of FAK and VCL ingastric mucosal in rats were observed by immunofluorescence method.Lastly, in orderto explore the role of the adhesion junction in the pathogenesis of stress stress ulcerand the preventive effect of hemostatic gel in stress ulcer,the rats were detectd EGF,IL-2, PAF level in serum and VCL, FAK level in gastric mucosa, after givenhemostatic gel by garaging. This study has great significance to the pathogenesis andprevention of stress ulcer.Methods:Based on the principle of sleep deprivation, animal models of stress ulcer in ratswere set up with the small platform over water method. By ELISA method,the level of EGF, IL-2and PAF in serum in rats were detected. By immunofluorescence method,the expression of VCL and FAK in gastric mucosa in rats were explored. Hemostaticgel was given by garaged.Results:1. The gastric mucosa expressed as varying degrees of erosion, ulcers, bleeding inevery stress group, Especially in stress5days and7days group, it can be seen moreserious mucosal injury. As the stress time extending, the UI was gradually increased（P<0.05）, EGF, IL-2level were decreased（P<0.05）, PAF level with stress severityincreased （P<0.05）.2. The expression of the gastric mucosa VCL and FAK show a downward trend（P<0.05）, as the stress time extending, the fluorescence photometric value of VCLand FAK were negatively correlated with the ulcer index.3. Given hemostatic gel, gastric ulcer index decreased significantly comparedwith the simple stress group（P<0.05）. The level EGF and IL-2were significantlyelevated in hemostatic gel groups compared with the stress group (P <0.05), and thelevel PAF was decline in stress group compared with the stress group (P <0.05).The expression of the gastric mucosa VCL and FAK were increased compared withstress group increased (P <0.05).ConclusionThe small platform over water method can be successfully constructed stress ulcermodel. There was a close relationship between VCL,FAK and stress ulcer, so itsuggested that adhesion junction is related with the occurrence of stress ulcer. It alsoconcluded that hemostatic gel had a preventive effect on the occurrence of stress ulcer. |
