| Background and Objective Primary carcinoma of the gallbladder (PCG) is acommon malignancy of the biliary system, in recent years, the incidence has increased yearby year. Annexin A1(ANXA1) was highly expressed in a variety of malignant tumors, andtumor cell proliferation, anoikis, invasion and metastasis of malignant behavior. P19protein isa tumor suppressor gene, can specifically inhibit the kinase activity of G1CDK4/6, to preventthe cells from G1to S phase transition, and cell cycle regulation, proliferation, apoptosis,migration, invasion and metastasis, and angiogenesisclosely related.Detection of ANXA1,P19genes expression in the gallbladder, gallbladder benign tissue. Explore the relationshipbetween the occurrence of the development of the gallbladder and gallbladder cancer clinicalpathological parameters. For further research and discussion on the origin and development ofgallbladder related genes, finding the effective method of diagnosis and treatment has certainclinical value and theory significance.Methods1. Collected from January2006to July2011, the General Hospital ofNingxia Medical Hepatobiliary Surgery surgical treatment and clinical, pathologic, andfollow-up data of56cases of complete gallbladder carcinoma, paraffin-embedded specimens,also collected over the same period due to cholecystitis or chroniccholecystitis benign diseasecholecystectomy specimens of56cases. By immunohistochemistry S-P method to detect ofANXA1and P19protein expression analysis of experimental results using SPSS11.5statistical software, sample rate compare with the the fourfold table or line×list chi-squaretest, correlation analysis of two-factor correlation with pearson. By quantitative Western-blot analysis of30cases of gallbladder carcinoma and30cases of non-neoplastic gallbladdertissues of ANXA1P19protein expression analysis of the results of x sgroups werecompared using the t test SPSS11.5statistical software line.Results1.Immunohistochemistry S-P method detected56cases of gallbladdercarcinoma and56cases of of benign gallbladder organization ANXA1positive expressionrate of69.64%(39/56),12.50%(7/56), Pl9positive expression rates were25.00%(14/56),83.93%(47/56), the chi-square test were statistically significant; ANXA1and P19expressioncorrelated with gallbladder patient’s gender, age, tumor size, and jaundice index was nosignificant relationship with tumor grading, TNM stage and lymph node metastasis-related;ANXA1and P19protein expression in the gallbladder was a negative correlation (r=-0.336).2.Western-blot quantitative analysis of30cases of gallbladder cancer tissue and30cases ofbenign gallbladder tissue the ANXA1protein grayscale scan and an internal reference β-actinprotein grayscale scanning the relative ratio of the average of were0.7442±0.0528,0.1570±0.0326,significant difference in the two sets of data by t test; P19protein grayscale scanningwithin the two organizations reference to the relative ratio of the average value of the β-actinprotein grayscale scanning were0.0976±0.0299,0.7299±0.0874, two sets of data by thet-test statistical difference.Conclusion1.ANXA1in the the PCG organization’s positive expression rate thehigher the degree of malignancy is the higher, the worse the prognosis of patients with theshorter survival.2. P19protein expression PCG organization the higher the rate, the higherthe degree of malignancy of the tumor, the worse the prognosis of the patient’s survival periodis the shorter.3. ANXA1&P19protein expression in PCG tissue and benign tissue in asignificant difference in the positive expression rate and tumor grading, TNM stage, lymphnode metastasis, can be used as indicators of the judgment development of PCG Clinical pathology reference.4. Joint detection the ANXA1&P19protein is expected to become animportant reference index for the PCG early diagnosis and prognosis.5. ANXA1and P19protein expression were independent factors affecting the prognosis of gallbladder carcinoma,between the two factors was negatively correlated. |
