[Objective]To investigate the expression and the effect of IL-17and TNF-αin lumbar intervertebral disc,to study the association betweent theeffect of IL-17and TNF-αin the degenearted lunber intervertebral disc;To supply the theoretical support for treating the degenerative discdisease and studying the cause of it.[Methods]The experiment group consisted of40cases of the fresh pulpy nucleustissue which were obtained from patients with lumbar disc herniation bysurgical intervention(18external anulus rupture,22external anulusintact),while10cases of the flesh pulpy nucleus tissue were harvestedBy operation for fracture of lumbar vertebrae acted as control group.ThemRNA and protein expression of IL-17and TNF-α in50specimens weredetected by semi-quantitative RT-PCR and immunohistochemicalstaining.Then,the results were analysed statistically.[Results]The mean IL-17and TNF-α protein and mRNA levels in the anulusfibrosus disrupted group and anulus fibrosus undismpted group are bothhigher than the control group.Between the lumbar intervertebral discof experiment group,the expression of the IL-17and TNF-α in the anulusfibrosus disrupted group is higher than that in anulus fibrosusundisrupted group(P<0.05).There was certain positive correlation between the mRNA and protein expression of IL-17and TNF-α inexperimental group(r=0.957,P<0.05)[Conclusions]IL-17and TNF-α in the cells of human lumbar intervertebral discmay be involved in the progress of human lumbar intervertebral discdegeneration.There was certain positive correlation be tween theexpression of IL-17and TNF-α. |
