Objective:To study the acute spinal cord injury in rats in the model, the injuryearly after a subdural injection Butin, observe the spinal cord organization ofneutrophils infiltrating the hind legs and the influence of the movement function.Materials and Methods:1. Choose only the growth of60health of march female SD rats (weighs about180-200g) randomly divided into three groups, group A (JiaShouShu group), asepticconditions only to rat T12do lamina resection, will spinal cord exposure, and not to anydisposal, directly will cut suture, the other two groups were improved with Allen ’sstrike law will rat spinal cord injury; Group B (contrast the treatment group) use certainexternal force of spinal cord damage potential energy exposure to T12organization,after the injury will be10μ l1%DMSO saline through a subdural injection into thedamage area; Group C (Butin treatment group) also damage the full force of potentialenergy exposure to T12spinal cord tissues, after the injury will be10μ l Butin solutionthrough a subdural injection into the damaged area. Rats after24h after with250mlsaline infusion, choose to damage area as the center, take up and down about3cmspinal cord tissue as specimens, extraction protein. Using the Western blot test analysisand pulp peroxidase (myeloperoxidase, MPO) active detection specimens. Rats feedingto2weeks after, record the rat movement the hind legs of change, and movementfunction analysis. Observation time points respectively is:1d,3d,7d, and d.2. This experiment according to records’s hind legs recovery change, the BBBrating method, the three groups of experimental rats comparison test.3. The rats24h after pouring specimens in the Western blot test analysis method, record every rats, spinal cord sample of the nf-kappa B p65and Caspase-3expression,and image analysis calculation system calculation group protein IOD sample value, andthen statistical analysis.4. Rats24h after pouring specimens, use ultraviolet visible spectrophotometerdetection protein samples OD value, according to the formula to calculate the activity ofMPO and statistical analysis.5. Statistics processing: experimental data to say, the use of SPSS13.0packagesingle factor analysis, with P <0.05said with a statistical significance.Results:1. The BBB score three groups of sample comparison, no damage in group A ofthe hind legs no change, compared with group A, B, C two groups of spinal cord injuryblow by gravity organization, the hind legs movement function is getting trend, summary record results from7d beginning, group C score is significantly increased thegroup B,7.61+/-0.59, A statistically significant difference (P <0.05).2. Through the Western blot test method of detection three groups of proteinsamples found, in group A the nf-kappa B p65and Caspase-3are in lower expression, Bgroup were significantly increased both expression, and after treatment for the group Cin which two group B protein expression is significantly reduced, finally through theIOD calculated value more than A statistically significant difference (P <0.05).3. Detection three specimens of protein OD value, analysis MPO activity, foundthat after24h SCI group B MPO active highest, after the group C Butin after treatmentof MPO activity of group A is high, and group C group B is A decreasing, calculationanalysis are statistically significant difference (P <0.05).Conclusions:1, Butin rat body movement function after SCI has improved role.2, Butin can restrain rats after SCI nf-kappa B p65excessive activation, reduceinflammation immune response, restrain Caspase-3expression, reduce cell apoptosis.
