Objective:To investigate the expressions of toll-like receptor4(TLR4) on circulating monocytes, The monocyte chemoattractant protein-1(MCP-1) in patients with acute coronary syndrome(ACS), and to explore the relationships between TLR4and MCP-1and coronary arteriosclerosis disease(CAD).Methods:25patients with Acute myocardial infarction(AMI),25patients with Unstable angina(UA) and13patients with stable angina(SA) confirmed by coronary angiography and30controls with a chest pain but no coronary heart disease confirmed by coronary angiography were recruited in this study. Expressions of TLR4on circulating monocytes from all patients were compared with flow-cytometry and MCP-1with enzyme-linked immunosorbent assay (ELISA). Analysis and compared measurement data with methods of mean±standard deviation, ANOVA test, Pearson correlation analysis et al; using SPSS13.0software package to analysis, with statistical significance when P<0.05.Results:①Detected by flow-cytometry,the expressions of TLR4on circulating monocytes were76.56±6.32in AMI,73.70±7.67in UA,63.20±6.86in SA and54.20±9.34in Sham respectively. The TLR4in the ACS was significantly higher than that in the SA(P<0.05) and patients with Sham group (P<0.01). The TLR4in the SA was higher than that in the Sham(P<0.05). the expressions of MCP-1were161.52±40.30in AMI,156.63±34.10in UA,141.32±29.26in SA and125.20±20.75in Sham respectively. The MCP-1in the ACS was significantly higher than that in the SA(P<0.05) and patients with Sham group (P<0.01). The MCP-1in the SA was higher than that in the Sham(P<0.05).②The level of MCP-1was positive correlated with TLR4in ACS (r=0.876, P<0.01).Conclusion:TLR4mediated inflammatory and immune mechanism involved in the occurrence and development of CAD.TLR4and MCP-1were related with plaque instability.
