However, some of its biological behavior, especially in tissue invasion and angiogenesis, are similar with malignant. At present, surgery is the best means to clear the EMT lesions. But small lesions, specifically microscopic lesions are often omitted from. Often require surgery and drug combination therapy for recurrent cases, especially deep infiltrating endometriosis is the difficulty in the treatment of endometriosis.The relapse treatment is more difficult. Vascular endothelial growth factor (VEGF), is widely recognized as the most important pro-angiogenic factor.Tumor necrosis factor-a (TNF-a) can also be synergistic VEGF to promote angiogenesis, and they jointly promote the ectopic endometrium planting, growth. This study observed intraperitoneal cisplatin infusion before and after treatment of endometriosis (EMT) in rat peritoneal fluid concentration of VEGF and TNF-a to explore the effect of cisplatin to inhibit the growth of endometriosis lesions,which uterineclinical drug treatment of endometriosis, especially in reducing the EMT’s invasion behavior, prevention of recurrence, to provide new ideas.Content:Vernon autologous transplant method was applied to establish the animal model of SD rat uterus endometriosis. EMT rats were intraperitoneal infusion appling cisplatin, while established two controls,they were0.9%sodium chloride solution infusion group and model control group. Took the concentration of VEGF and TNF-a in the peritoneal fluid as observed index, to evaluate the effect of intraperitoneal cisplatin perfusion in inhibiting the growth of endometriosis lesions.Method:Used of clean, healthy, female, sexual maturity, no mating SD rats (45) as experimental subjects to establish EMT rats model (model group). The EMT rats were divided randomly into three groups:Cisplatin celiac perfusion group (model group B), physiological saline celiac perfusion group (model group C)and model control group (model group D). Sham-operated rats, only pulling the uterus as normal control group (control group A).4weeks after surgical, we opened the abdominal cavity again,observed the growth of implanted endometrial and collected samples. EMT rats were divided randomly into three groups that were deal with different methods. Cisplatin celiac perfusion group were given Cisplatin2mg/kg+distilled water2ml by peritoneal lavage. Physiological saline celiac perfusion group were given a large number of0.9%sodium chloride solution (10ml) by peritoneal perfusion. The model control group were not given treatment after collected samples. Groups were fed conventionally and were intramuscular injected of antibiotics to prevent infection after surgery.4weeks later, opened the abdominal cavity, observed the growth of implanted endometrial and collected peritoneal fluid samples.We determinated the concentration of VEGF and TNF-α in the peritoneal fluid of rats of each group by Enzyme Linked-Immuno-Sorbent Assay(ELISA),and then we tested the correlation between VEGF and TNF-a.Results:The concentration of VEGF and TNF-a in the peritoneal fluid of the EMT rats was significantly higher than that of normal group, differences were statistically significant(P<0.05);After the different treatment, the concentration of VEGF and TNF-α was significantly lower in the Cisplatin celiac perfusion group than that of the other groups, differences were statistically significant(P<0.05). The concentration of VEGF and TNF-α was significantly correlated before and after treatment (P<0.01).Conclusions:It was found that the concentration of VEGF and TNF-α in the peritoneal fluid of the EMT rats was significantly higher than that of normal rats. Cisplatin celiac perfusion therapy could reduce the concentration of VEGF and TNF-α in the peritoneal fluid,could suppress ectopic endometrial growth and could make ectopic endometrial to be shrinking. VEGF and TNF-α may play a certain role in the occurrence and development process of endometriosis,and there was a positive correlation between the VEGF and TNF-α.
