Objective:To study the changes of trace elements between normal children and children with acute lymphocytic leukemia (ALL) during different cycles and high-dose methotrexate (HDMTX) chemotherapy.Methods:The vein blood samples were collected from20normal children,20children diagnosed primarily as ALL and40children with ALL before and after high-dose methotrexate (HDMTX) treatment including4children of the first,8children of the second and8children of the third cycle HDMTX treatment. In isolated plasma, the plasma concentrations of zinc, iron, copper, manganese, chromium, aluminum and lead were measured with inductively coupled plasma atomic emission spectrometry (ICP-AES) technique. Children with ALL were divided into All ALL group, ALL group treated primarily, ALL group of the complete remission (before HDMTX treatment) and after HDMTX treatment.Results:1.There were no significant difference about the plasma concentrations of zinc, iron, copper, manganese, chromium, aluminum and lead between the male and female normal children group as well as between3-7years old group and7-12years old group (P>0.05)2.All ALL group and ALL group treated primarily compared with normal children. The plasma concentrations of zinc、iron、chromium were respectively lower, and copper、lead were respectively higher(P<0.05). But the plasma concentration of manganese was lower, aluminum was higher(P>0.05). ALL group of the complete remission compared with normal children. There were no significant difference about the plasma concentration of zinc,iron,copper, manganese, chromium, aluminum and lead (P>0.05).3. ALL group treated primarily compared with ALL group of the complete remission.In ALL group of the complete remission, the plasma concentrations of zinc、 iron、chromium were respectively higher, and the plasma concentrations of copper、lead were respectively lower (P<0.05). But the plasma concentration of manganese was higher, and aluminum was lower(P>0.05).4. During the first, second and third cycle of HDMTX treatment after ALL of complete remission, there were no significant difference about the plasma concentrations of zinc, iron, copper, manganese, chromium, aluminum and lead (P>0.05).5.Children with ALL before HDMTX treatment compared with children after HDMTX treatment. The plasma concentrations of zinc, iron, copper and chromium were respectively lower (P<0.05).But the plasma concentrations of manganese、aluminum and lead were lower (P>0.05).Conclusions:The concentrations of trace elements had apparent changes in children with ALL,especially ones diagnosed primarily. With the disease remission, the concentrations of trace elements had a tend to be corrected. The concentrations of trace elements would temporarily decline in children with ALL during the first, second and third cycle of HDMTX treatment after ALL complete remission, but the concentrations of trace elements can go back the last normal levels before next HDMTX treatment. It is suggested that there are the effect of ALL and HDMTX chemotherapy on the contents of trace elements in children with ALL.
