Technology drives the progress of humanity。In recent years, with the rapid development of science and technology, medicine has made great progress and breakthrough, but for cancer treatment is still unable to attack, and tumor incidence and mortality rates are rising rapidly.Although treatment technology had great progress and improve, but5years of survival rate is still not improved, investigate its main reason is when most patients see a doctor has been late, lost the best treatment opportunity.Lung cancer is no exception, of course, is still the incidence and mortality in the lead of the malignant tumor in the world within the scope, and this highest among all the malignant tumor about85%are non-small cell lung cancer (NSCLC).Thus the study of early diagnosis and treatment of lung cancer especially non-small cell lung cancer (NSCLC) to become the world’s one of the main goals of the medical profession have been exploring.Among gene therapy has become the most popular research approaches,Because a lot of research results show that this approach is expected to become the effective way of treatment for lung cancer.And STK33is role ever found in the latest non cancer gene,Located in human chromosomes11p15.3area.But for many of the tumor,The main reason is that it happened is because of some special genes of dysfunction which is correlation of the tumor,the study found that the STK33in chromosomal regions of multiple genes associated with the occurrence and development of clinical diseases.And research shows it also are expressed in the in many other tumor tissue.For this reason this research through the Trans-well technology of STK33in lung cancer cells after gene silencing hit number and distance, and utilize RT-PCR and western-blot to test the integrin mRNA and protein which In STK33siRNA-2 group and contrast the different expression in normal lung cancer cell group.Discuss the influence of STK33in transfer of lung cancer cells.objective:Analysis the discretion of the STK33that expression in lung cancer cells,and effects on lung cancer cells invade ability.Discuss STK33on membrane surface protein expression related to tumor metastasis,discuss STK33on membrane surface protein expression related to tumor metastasis (such as integrin),to study whether STK33has certain effects on the transfer of lung cancer cells.Methods:Discuss STK33on membrane surface protein expression related to tumor metastasis,discuss STK33on membrane surface protein expression related to tumor metastasis (such as integrin),through the RT-PCR and western-blot to study whether STK33has certain effects on the transfer of lung cancer cells.Results:1.Technical inspection of the Trans-well to study STK33siRNA-2group the number of lung cancer cell migration and the migration distance are less than the control group of normal lung cancer cell/There are significant differences (P=0.000).2. RT-PCR and western-blot to test the integrin mRNA and protein STK33siRNA-2group are less than the control group of normal lung cancer cell,There are significant differences (P=0.000)Conclusions:STK33gene and lung cancer cell invasion have a close correlation,Possible become a new hope for lung cancer treatment.
