Shou Tai Wan (STW) is a traditional Chinese herbal prescription, which is composed of Dodder, Teasel, Viscum album and Colla corii asini. It’s medicine value is regulating immunity and internal secretion, inhibiting uterine smooth muscle contraction and strengthening pituitary-ovary luteinizing. Now, due to the effective cure with minimum side effects, STW is widely used to in pregnancy to tonify kidney for preventing miscarriage and treat habitual abortion. Researching the content in Shou Tai Wan decoction is helpful to explore the tocolysis material foundation of Shoutaiwan from the nutritional point, make this prescription more efficient to benefit patients.1. To establish a method to simultaneously determine trace element contents in Shou Tai Wan. Concentrations of elements were determined by ICP-AES. We also study the dissolution of trace elements in the Shou Tai Wan decoction. Dodder, Teasel, Viscum album and Colla corii asini were digested by Nitric acid-hydrogen peroxide system, decoction were digested by Nitric acid-perchloric acid. The contents and solubility of the trace elements in Shou Tai Wan herbal medicine and decoction were compared. Experimental results show that, the recovery rates of the procedure were between80.7%and117.0%, the RSDs were between0.75%and7.06%. ICP-AES combining the microwave digestion technology was simple, rapid, precise, convenient and suitable for determining trace elements of traditional Chinese medicine. Shou Tai Wan decoction contains12kinds of elements of K, Na, Ca, Mg, P, Al, Fe, Mn, Zn, Cu, Co, Cr, etc. As and Li were not found. The dissolution rate of K, Na, P, Mg, Mn and Co were relatively high.2. A simple, selective, rapid and reliable method for determination of selenium in Shou Tai Wan by atomic fluorescence spectrometric has been developed. Comparison was made between the contents and dissolution rate of selenium in Shou Tai Wan herbs and decoctions. The herbs and decoctions both contain selenium, the selenium content in Dipsacus herb is higher than other herbs, however, selenium dissolution rate in Dodder decoction is higer than other decoctions, but lower than the Shou Tai Wan decoction. The average recovery of Se is in the range of88.8%-97.1%, with RSDs of1.8%-2.6%. The present method was successfully applied to detect selenium in Chinese herbal compound of Shou Tai Wan.3. The use of anion-exchange chromatography with integrated pulsed amperometric detection allows good separations of18amino acids in Shou Tai Wan decoctions. SSDBX solid-phase extraction cartridge was use to purify amino acids, optimizing the extraction process. This method avoids pre or post-column derivatization, which increases the method rapidity and simplicity, and reduces sample manipulation. It is shown from the results that is a sample, direct and practical method suitable for simultaneous determination of18amino acids in Shou-Tai-Wan decoction. Under optimized conditions, this method was applied to determination of amino acids in Shou Tai Wan with satisfactory results. The detection limits for the analytes ranged from0.07-5.00ng/L. The linearities for all analytes were over two orders of magnitude with the correlation coefficients greater than0.995(except for asparagine). The recovery rates were in the range of71.2%-120.9%.4. A extraction and purification method based on solid-phase extraction(SPE) was used in traditional Chinese medicine Shou Tai Wan to analyze three flavonoids(Hyperoside, Quercetin and Kaempferol) with Ultra performance liquid chromatography. This paper discussed the flow-rate, mobile phase and the extraction condition. Good precision and spike recovery were observed using the optimized method for each of these complex samples. The recovery rates were in the range of90.0%-100.8%, with RSDs of1.4%-4.4%. It shows this method is a rapid, sample and sensitive method was detected for the analysis of flavonoids in traditional Chinese medicine Shou Tai Wan.
