Objective:To investigate the inhibitiory effect of JuanBi particle on collagen(CII)-induced arthritis (CIA) in mice and its relative mechanisms, to clarify the scientific basis of superior efficiency of Chinese medicine in the clinical treatment on Bi Syndrome (wind-cold-dampness Bi) according to Wu School-based on strengthening yang theory.Method:1.Delayed type hypersensitivity (DTH) model in mice induced by DNFB was established for testing the vivo immunosuppressive effects.2.Type II bovine collagen(CII)-induced arthritis (CIA) in DBA/1mice were established for investigating the antiarthritic potential of JuanBi particle, mice were divided into three groups including model CIA, MTX and JuanBi particle groups.The severity of arthritis was evaluated by the clinical score. The histological damage was assessed by histological examination (H&E) and Molybdenum target X-ray.The effects of JuanBi particle on Lymphocyte proliferative ability in collagen(CII)-induced arthritis were determined by [3H]-thymidine incorporation assay, the cytokine production levels of IFN-γ and IL-10in collagen(CII)-induced splenic lymphocytes were detected by ELISA.Results:1.JuanBi particle had inhibited action on the delayed type hypersensitivity (DTH)(p<0.05).2.The clinical score and inflammatory cells infiltration in CIA mice were significantly reduced in the JuanBi particle administration group (p<0.01). the CII-specific immune responses has been suppressed by the JuanBi particle (p<0.01), the produce level of IL-10in collagen(CⅡ)-induced splenic lymphocytes (P<0.05) were increased, while the IFN-γ has no significant changed.Conclusion:1. JuanBi particle the vivo immunosuppressive effects.2.The results indicated that JuanBi particle can suppress the polyaritis in CIA mice, the mechanisms may relate to promote the cytokine production levels of IL10and to mediate negative regulation.
