The Compound Juanbi Granular Treatment Of Rheumatoid Arthritis Pharmacodynamic And Acute Toxicity Test Research

Objective: To study the effects of Juanbi particles in treating (Rheumatoid Arthritis,RA).RA is a common clinical difficult disease,which rate of causing disablility is high,so it influence the patient's life quality directly.In treating ,there is still not a ideal effectual medicine with little toxin side effect at present.RA belongs to"be syndrome"in TCM ,which has unique understandings of the etiology and pathogeneticology of RA and has created a lot of effectual prescriptions in practice.We choose Chinese herbal medicines .JuanBi granules has the function of clearing away expelling wind and removing damness, dispel cold and relieve pain. For the treatment of RA are there anti-inflammatory, analgesia ,regulating immune function role of pharmacology. And to investigate the mechanism of the treatment of RA.Methods: (1)The object of this study is mice ,determine the maximum capacity of mice orally of Juanbi granule's extract(.2)observe the anti-inflammatory effects of Juanbi granules,By using xylene induced ear edema in experimental studies (3)The object of this study is mice, and observe the analgesic effects of Juanbi granules ,by using acetic acid writhing method.(4)Take the adjuvant induced arthritis rats as models,make the degree of Rat's paw swelling, Spleen and thymus weight index,and the Pathological changes of Synovial as the Index of Detection and change of Serum inflammatory factors.Research the Juanbi particles's effect of express of adhesion molecule about the related factor of Synovial tissue. Explore the effect and the mechanism ,which is about anti-inflammatory and immunoregulation ofCompound Juanbi of granules in rats.Result: The oral administration experiments with the largest Volume indicate that, the maximum capacity of oral administration with Juanbi compound granules of the mice is 248g crude drug / Kg / day,which is 179.7 times of the Daily oral dose for adults , (Adult body weight calculated according to 70kg), For the pharmacodynamics of high doses of 4.92-fold in mice, ( the high dose of daily oral drug of pharmacodynamics of Mouse with 50.4g/kg).The Pharmacodynamic studies show that the Compound of Juanbi granules have effects of anti-inflammation, analgesic ,and immune regulation. It also can supress the edema of AA rat paw, and improve index of immune organs ,such as spleen and thymus, they are in AA rats. Meanwhile,it can significantly improving the inflammation and hyperplasia of the joint of synovitis in AA rats , the Compound of Juanbi inhibited high number of inflammatory cytokines ,such as IL-1β, TNF-a secretion in AA rats , In the partial synovium in joint,the Juanbi granules can supress the excessive express of abnormal ICAM-I in Synovial tissue Conclusion: The Compound of Juanbi granules has the effect of analgesic, anti-inflammation, and immune regulation. The effects of anti-inflammation and immune regulation of Juanbi granules are related to the inhibition of excessive inflammatory cytokine secretion; The Juanbi granule's supress the excessive secrete of adhesion factor may be the basis of Pathology.which controls the inflammation of Synovium and forbid the formation of Pannus.
