Seven Viral Pathogens Infections In Children With Diarrhea Disease

Objective: To investigate the status of adenovirus(AdV)、 rotavirus(RV)、 aichivirus(AI)、 calici virus(HuCV)、 parechovirus(HPeVs)、 enterovirus(EV) andastrovirus(AstV) in children in5-years old with diarrhea,and study the epidemiologicalcharacteristics of AdV. Methods:315fecal samples were collected from both in-patientand out-patient children with diarrhea from January to December of2011in Urumqi. Thenthe nucleic acid(DNA) was extracted from all315samples.After that, AdV was detectedby polomerase chain reaction(PCR).All of the positive samples were sequenced andcompared with other AdV in GenBank by Blast and DNAstar. RNA was Extracted fromall315fecal samples. Then it was changed into cDNA by reverse transcription.At last allthe cDNAs were amplified and the other six viral pathogens were detected by multiplexreverse transcription polymerase chain reaction (RT-PCR).The three positive ampliconsselected from all positive samples were sequenced to confirm the results. Results: Sixvirus pathogens were detected exceptAstV, and the detection rate was55.24%(174/315).The most importaint viral pathogen was RV with the detection rate27.30%(86/315), andAI, HPeVs, HuCV,EV and AstV were18.41%(58/315)、12.38%(39/315)、12.06%(38/315)、5.08%(16/315) respectively,while the cocurrent infections occupied17.14(54/315). All the12strains shared73%~99%nucleotide sequence identity withdifferent reference strains. There were no statistically significant differences in sex andnation in all the six viral pathogens. RV infection occurred mainly in autumn andwinter(September to October and January), accounted for48.84%(42/86),AI、HpeVs andEV it was occurred mainly in July to August、August to September and June to October,accounted for55.17%(32/58)、53.85%(21/39) and100.00%(16/16) respectly.And theHuCV and AdV infections had no seasonal flunctuation. The age of AdV infections wasmainly in seven to twelve months,accounts for75.00%(9/12),AI was less than twenty-fourmonths,accounts for87.93%(51/58),and other four viral pathogens had no difference onthe frequencie(P>0.05). The main subtypes of AdV were enteric adenovirus (subgenus F)Ad41with a positive rate of2.54%(8/315),followed by non-enteric adenovirus (Ad1, Ad3,Ad5,Ad7,Ad31) with a positive rate of1.27%(4/315),and Ad40was notdetected.Conclusions: RV was the most important cause of infant diarrhea inUrmuqi,China,the role of AI、HPeVs、HuCV、EV and AdV infection can not be ignoredin the infantile diarrhea.
