| With the improvement of people’s living standard, the morbidity of obesity and type2diabetes increases constantly and threatens human health seriously. Since the occurrence and development of type2diabetes is closely related to gut microbes, research in this field has been a focus among society and medical professionals.In the experiment, gut microbial community of two genotypes mice, spontaneous type2diabetes KKAy mice and normal C57BL/6J mice, are analyzed by PCR-DGGE, similarity coefficient (C5), cloned bands sequencing, gene libraries comparison, phylogenetic analysis, etc. Statistical results show that the gut microbial community between two genotypes mice clearly distinct. Cs is only26%-44%among different genotypes, while Cs is82%-100%among the same genotype. Furthermore, the cloned and sequenced results of thirteen dominant bands from DGGE profiles exhibit more differences on gut microbial structure. All sequences are compared in GenBank and RDP database and a phylogenetic tree is developed. The gut microbes, which are associated with diabetes, are identified and their physiological functions are interpreted.The study of the influence of drug on gut microbes in KKAy mice is described. Faecal samples are collected in5different points of time during the drug experiment and PCR-DGGE is run. Changes of gut microbial diversity are determined and principal component analysis (PCA) is conducted. During the experiment, gut microbial diversity of normal group and high-Chinese-medicine-dose group mice remains high and is more stable, which of low-Chinese-medicine-dose group is higher than which of model control group and Pioglitazone group. And that of Pioglitazone group falls at the highest rate. The influence of Pioglitazone on gut microbes is unidirectional in PCA, but Yi Qi Hua zhuo Capsules is polydirectional. Results show that the effect of Yi Qi Hua zhuo Capsules is different with Pioglitazone, the former having an advantage of maintaining microbial diversity and improving gut microbial structure.We compares the quantity of Bacteroides and Lactobacillus in each group at different time points by real-time fluorescence quantitative PCR. Bacteroides quantity in both high and low-dose of Chinese medicine groups falls close to normal group level30days after drug. But model control group and Pioglitazone group do not change and still remain at a higher level. Lactobacillus quantity in model control group falls during the experiment, but that of the rest groups rise significantly. The result explains that Yi Qi Hua zhuo Capsules has special regulation functions for Bacteroides quantity, and increasing quantity of Lactobacillus can alleviate condition of type2diabetes.Gut microbes content many biological mysteries and mankind needs more and more advanced technology to decipher the secrets. From our study, we believe that Chinese medicine has a very broad and effective use in treatment of type2diabetes, and deserve more attentions in the science field. |
