[Objective]To observe change regularity of the expression of Netrin-1and its receptors in lung carcinoma, expecting to provide a testing basis for the diagnosis, treatment and prognosis of lung carcinoma.[Methods]In this study, to construct tissue microarray of lung cancer, we collected80lung carcinoma tissues as experimental group and20normal adjacent lung tissues as control group.The expression states of Netrin-1, DCC and UNC5H protein in lung cancer and adjacent tissues were determined by immunohistochemical staining, and Terminal-deoxynucleotidyl Transferase Mediated dUTP Nick End Labeling(TUNEL) was used to detect the apoptosis index AI of cells in the lung carcinoma tissues, in order to analyze the relation between Netrin-1and its receptors and clinical pathologic characteristic as well as cell apoptosis.[Results]1. The positive expression rate of Netrin-1in the lung tissues was56.25%, and the positive expression rate of it in normal adjacent lung tissues was5.00%; statistically significant difference was found between the expression of Netrin-1in lung cancer tissues and their normal adjacent lung tissues(χ2=16.918,P<0.05). 2. The positive expression rate of DCC and UNC5H in the lung tissues respectively was22.50%and13.75%, and the positive expression rate of them in normal adjacent lung tissues respectively was80.00%and65.00%; statistically significant difference was found between the expression of DCC in lung cancer tissues and their normal adjacent lung tissues(x2=23.574, P<0.05), statistically significant difference was also found between the expression of UNC5H in lung cancer tissues and their normal adjacent lung tissues(χ2=23.040,P<0.05).3. The protein expression of Netrin-1and DCC in lung cancer tissues was interrelated to histological type and degree of differentiation(P<0.05), but not related to sex, age, tumor size and lymph node metastasis (P>0.05); statistically significant difference was not found between the expression of UNC5H and clinical pathologic characteristic(P>0.05).4. The expression of protein Netrin-1, DCC and UNC5H in lung cancer tissues was analyzed, there was negative relation between the expression of protein Netrin-1and DCC(r=-0.309, P<0.01), but no relation between the expression of protein Netrin-1and UNC5H, as well as the expression of protein DCC and UNC5H (r=0.133, P>0.05; r=0.046, P>0.05).5. In lung cancer tissues, the apoptosis index in the group of Netrin-1positive expression was significantly lower than in the group of Netrin-1negative expression, and the apoptosis indexes in the group of DCC and UNC5H positive expression were significantly higher than in the group of DCC and UNC5H negative expression (t=7.983, P<0.05; t=9.612, P<0.05; t=9.977, P<0.05). The apoptosis indexes of different expression levels of protein Netrin-1, DCC in lung carcinoma were analyzed, the difference was statistically significant (F=15.875, P<0.05). The results also showed that the apoptosis index in the group of Netrin-1(+)+DCC (-) was significantly lower than in the group of Netrin-1(+)+DCC (+), and the apoptosis index in the group of Netrin-1(+)+DCC (-) was also significantly lower than in the group of Netrin-1(-)+DCC (-)(t=2.191, P<0.05; t=2.660, P<0.05).[Conclusion] 1. A higher expression of Netrin-1was detected in lung cancer tissues but a lower expression of it in normal adjacent tissues, and a lower expression of DCC was detected in lung cancer tissues but a higher expression of it in normal adjacent tissues; both of the expression of Netrin-1and DCC were associated with histological type and degree of differentiation, suggesting that excessive expression of Netrin-1and loss of expression of DCC may relate to the occurrence and development of lung carcinoma.2. A lower expression of UNC5H was detected in lung cancer tissues but a higher expression of it in normal adjacent tissues, it indicated that UNC5H may have characteristic of a tumor suppressor gene; and there was no relationship between the expression of UNC5H and clinical pathologic characteristic.3. There was a negative correlation between the excessive expression of Netrin-1and loss of expression of DCC in lung cancer tissues, this showed that the protein Netrin-1and DCC may play a role together in occurrence and development of lung carcinoma. The expression of Netrin-1protein and DCC protein was detected at the same time could make for judging the prognosis of lung carcinoma.4. In lung cancer tissues, the apoptosis index in the group of Netrin-1positive expression was lower than in the group of Netrin-1negative expression, suggesting that Netrin-1protein could inhibit apoptosis; the apoptosis indexes in the group of DCC and UNC5H positive expression were higher than in the group of DCC and UNC5H negative expression, suggesting that DCC and UNC5H protein could induce apoptosis; Netrin-1and DCC was abnormal at the same time, the apoptosis index in lung cancer decreased notably, which further confirmed the Netrin-1and DCC played a crucial role in lung cancer cell apoptosis and jointly participated in the occurrence of lung cancer through the regulation of cell apoptosis.
