| Objective To investigate the effects of Prostaglandin E1on liver cellapoptosis of rats with obstructive jaundice (OJ) and the correlation of multidrugresistance protein2(MRP2).Methods72healthy male SD rats were average divided into three groups atrandom:the control group (CG group); the common bile duct ligation group (OJgroup); the common bile duct ligation and under PGE1treatment group (OJ+PEgroup). And to establish OJ model later. Each group were divided into three littlegroups by postoperation time:3days;7days,10days. All groups were collected acertain amount of inferior vena venous bloods and part of the liver tissue to measuredthe serum liver function level.HE staining to observing the liver pathologymorphology change, TUNEL method to determinating the liver cell apoptosis index(AI), immunohistochemical staining to determinating MRP2protein expression andRT-PCR to measuring its mRNA level.Results①Compared with CG group, OJ group’s rats serum aminotransferase andbilirubin level increased (P <0.05), the liver tissue pathology change obviouslyaggravated, liver cell AI increased (P <0.05), MRP2protein expression reduced (P <0.05),and its mRNA expression down. With the time of common bile duct ligationextending,the changes are more obvious.②Compared with OJ group, serumaminotransferase and bilirubin were lower in OJ+PGE1group.The liver tissuepathology changes ease; Liver cell AI reduced(P <0.05); MRP2protein and itsmRNA expression increased in each group(P <0.05).Conclusion (1)Rats after biliary obstruction can cause liver function damage and liver cell apoptosis increased significantly, liver cell apoptosis is one of the importantmechanisms in the liver injury caused by obstructive jaundice.(2)Prostaglandin E1can relieve to some extent the liver function damage caused byOJ in rats and inhibit liver cell apoptosis, increase MRP2protein expression may be isone of its mechanisms. |
