Studies On Bellidifolin To Neurorehabilitation After Sciatic Nerve Injury

Objective:To study whether the Bellidifolin have the rehabilitation effect to injuried sciatic nerve rat, to explore whether Ciliary Neurotrophic factor (CNTF) is involved in this mechanism, to make basic foundation for further investigation. Methods:Male wistar rats225to make sciatic nerve injuried model, with right sciatic nerver was cut and sewed under microscopy, left sciatic was sham. Rat devided into5groups randomizely, as control group, Bellidifolin25mg group,50mg group、75mg group and Mecobalamin group, each group were45rats. Control group just injected sodium chloride, others peritoneal injected different dose of Bellidifolin and Mecobalamin after operation. Detected foot print after1,3and5weeks, counted the Sciatic Funtion Index(SFI), Motor Nerve Conduction Velocities(MNVC), Triceps Surae Weight(TSW), immunochemical method analysed CNTF expressin. Results:(1)SFI results:1weeks after operation, control group, mecobalaming grop and Bellidifolin25mg group,50mg group, and75mg group were:-84.35±4.87,-45.20±2.30,-70.42±4.21,-57.73±3.46and-64.38±4.38, compared with control group, others had differences (P<0.05), Bellidifolin groups versus mecobalaming group, there were statistically differences(P<0.05), within Bellidifolin groups.50mg group had difference compared with25mg and75mg groups.(P<0.05)(2) the NMCV. The largest amplitud and TSW also showed there were differences among bellidifolin groups, control group and mecobalaming group. Even within Bellidifolin groups,50mg group compared with25mg and75mg groups, there were statistically differences(P<0.05).(3)3weeks compared each groups with Immunohistochemistry analysis, CNTF expression showed there were statictically differences among billidifolin50mg group and25mg,70mg groups, of50mg group are higher than others(P<0.05). Conclusions:(1) Bellidifolin can elevate the SFI valure of cut and sewed sciatic nerver, improve the motor nerver conduct velocity and the weight of triceps surae.(2)The concentration of Bellidifolin50mg has better effect than25mg and75mg.(3) Bellidifolin can improve the injuried sciatic rehabilitation, and CNTF is involved.
