Objective:To investigate the expression of heat shock protein70(HSP70) in human hepatocellular carcinoma (HCC) and its relationship with HCC differentiation grades, serum HBV markers, alpha-fetoprotein levels and mass diameters.Methods:During November2010to January2012, a total of38cancerous samples and their non-cancerous adjacent tissues were obtained from38patients with HCC who underwent surgical liver resection at Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology. Eight normal liver samples were collected from surgical patients with liver hemangioma as control group at the same period. Immunohistochemical staining and Western blotting were employed to detect the expressions of HSP70in the tumor tissues, tumor adjacent tissues and normal liver tissues (from adjacent tissue of hemangioma). The expressions at different tissues were quantified by Image-Pro Plus6.0and Gelpro32, and data were statistically analyzed with SSPS17.0.Result:1. The expressions of HSP70in cancerous tissues were higher than those in non-cancerous adjacent tissues and in adjacent liver tissues of hemangioma, the IOD values were26122.15±22242.04,16414.24±23037.43and5027.70±5173.02, respectively (P<0.05).2. The relationship between the expression of HSP70and tumor histological differentiation grade. The expression of HSP70in cancerious tissues tended to increased with degrees of histological differentiation reduced(P>0.05). Each of their IOD value was26524.56±25005.78and21236.94±18969.79and19664.95±11924.96(mean±SD). But in adjacent cancerous tissues, the trend was reversed. Their medians were7086.09,6399.70and7034.77. Interquartile range(IQR) of them were14118.49,10680.77and48156.53(P>0.05)3. The relationship was between expression of HSP70and serum hepatitis B virus (HBV) DNA. The expression of HSP70was lower in serum positive HBV DNA of cancerous tissues than negative HBV DNA of tissues(P<0.05). Each of their IOD values was19167.18±15790.88and31474.96±22420.76(mean±SD). The trend was reversed in adjacent cancerous tissues.The expression of HSP70in positive HBV DNA of adjacent cancerous tissues was little more than negative HBV DNA of adjacent tisssues. Each of their medians was6802.38and8298.05IQR was12732.08and19658.88(P>0.05), respectively.4. The relationship was between expression of HSP70and cirrhosis. There was no significant difference of HSP70in cirrhosis and non-cirrhosis(P>0.05). IOD values of cirrhosis and non-cirrhosis were24329.1663±20262.06and26203.3122±16335.49275(mean±SD). Each median of IOD in adjacent cancerous tissues of cirrhosis and non-cirrhosis was6802.38and12282.28. Each IQR was12269.11and31536.99(P>0.05)5. The relationship was between expression of HSP70and serum hepatitis B surface antigen (HBsAg). Compared positive HBsAg of cancerous tissues, HSP70expressed little more in negative HBsAg of cancerous tissues.P value was0.001between positive HBsAg of cancerous tissues and their adjacent tissues. Compared positive HBsAg of cancer and negative HBsAg of adjacent cancerous tissues, value P was0.01. Adjacent cancerous tissues of positive and negative HBsAg of P value was0.028. Each median of expression of HSP70in positive HBsAg cancerous tissues, adjacent their tissues, negative HBsAg cancerous tissues and adjacent tissues was21224.99,8797.38,21426.15and1605.86. Their IQR were32163.72,15877.19,23053.62and13280.32. 6. The relationship was between expression of HSP70and serum alpha-fetoprotein (AFP) levels. The expression of HSP70in serum AFP<400ng/ml of cancer was lower than AFP>400ng/ml of cancer (P<0.05). There were statistical significance between AFP<400ng/ml of cancer and adjacent tissues of AFP>400ng/ml and AFP<400ng/ml (P<0.01). Their medians were8897.73,6261.40,21539.67, and7941.92, respectively. IQR of them were40726.18,20511.11,27003.47and12574.71.7. The relationship was between HSP70and liver function. HSP70in normal liver function of cancer expressed more than its in dysfunction of liver cancer(P<0.05). Each of them was28071.02±23559.67and18276.48±14710.36(mean±SD). The medians of expression in adjacent tissues were7372.15and6833.43and their IQR were23489.6and11182.13in normal liver function and dysfunction of liver (P>0.05).8. The relationship was between HSP70and mass diameter. The expression of HSP70in mass diameter>5cm of cancers was significant difference from adjacent cancerous tissues(P1=0.007,P2=0.018). HSP70expressed in tumor size <5cm of cancers was more than adjacent cancerous tissues too(P3=0.031,P4=0.013). The medians of these tissues were21565.66,6833.43,18734.80and8298.05and their IQR were21565.66,6833.43,18734.80, and8298.05respectively.9. Western blotting result:HSP70IOD/Actin IOD in cancerous tissues, adjacent cancerous tissues and adjacent hemangioma of liver were2.57±2.01,1.25±0.81and0.80±0.36(mean±SD,P<0.05).Conclusion:HSP70was overexpressed in tumor tissue and correlated with grade malignancy of tumor, serum positive/negative HBV DNA, AFP levels, liver function and so on. HSP70may play a role in hepatocarcinogenesis and development of HCC. Further research should be essential to the significance of HSP70in HCC.
