Salivary adenoid cystic carcinoma is one of the most common epithelial malignant neoplasms, accounting for10%of salibary gland epithelial malignat tumors, It has the charcateristics of developing slowly, being highly infilitrative, and being likely to invade blood vessels and nerves. It is prone to recurrence and distant metastasis at early stage of the protential growth of tumor cell. Infiltration and metastasis are the main causes human of death. And the foundation of basic structure is tumor angiogenesis. Therefore, inhibiting angiogenesis has become the key of cancer treatment. The object of this study is to investigate the expressions of correlation between STAT3、VEGF、bFGF and MVD in Salivary adenoid cystic carcinoma, and to study the relationship between the clinic significance of the expressions of STAT3mRNA、VEGF mRNA and angiogenesis.50cases of adenoid cystic carcinoma of salivary gland and15cases of normal salivary gland as control are chosen in pathology department of the first affiliated hospital of the He bei North University. The expressions of STAT3mRNA and VEGF mRNA in salivary adenoid cystic carcinoma and normal salivary gland tissue are studied by in situ hybridization. The expressions of STAT3、VEGF、bFGF and CD34in salivary adenoid cystic carcinoma and normal salivary gland tissue are studied by immunohistochemistry.The microvesse density is counted through the microscope.The experimental results show:1The positive rate of STAT3and STAT3mRNA in salivary adenoid cystic carcinoma tissues are respectively66%(33/50)and72%. which are significantly higher than that of the normal group (P<0.05). The expressions of STAT3protein and STAT3mRNA are concerned with lymph node metastasis and TNM stage(P all<0.05), and are irrelevant with the gender and age (P all>0.05).2The positive rate of VEGF and VEGF mRNA in salivary adenoid cystic carcinoma tissues are respectively66%(33/50) and72%(36/50), which are significantly higher than that of the normal group (P<0.05). The expression of VEGF protein is concerned with lymph node metastasis histological type and TNM stage(P all<0.05), and is irrelevant with the gender, tumor site and age (P all>0.05); The expression of VEGF mRNA is concerned with lymph node metastasis and TNM stage(P all<0.05), and is irrelevant with the gender, histological type, tumor site and age (P all>0.05).3The positive rate of bFGF protein in Salivary adenoid cystic carcinoma tissues is66%(33/50), which is significantly higher than that of the normal group (P<0.05); The expression of bFGF protein is concerned with lymph node metastasis histological type and TNM stage(P all<0.05), and is irrelevant with the gender,tumor site and age (P all>0.05).4The mean MVD of salivary adenoid cystic carcinoma tissues is56.93±12.43, which is significantly higher than that of the normal group11.80±3.49(P <0.05). The mean MVD of salivary adenoid cystic carcinoma tissues is concerned with lymph node metastasis, distant metastasis and TNM stage(P all<0.05), and is irrelevant with the gender and age (P all>0.05).5The correlation between STAT3、VEGF、bFGF and MVD shows that positive correlation is found between STAT3and MVD in salivary adenoid cystic carcinoma(P<0.05). The mean MVD are correlated with the expression of VEGF closely (P<0.05). The mean MVD is closely correlated with the expression of bFGF(P<0.05), so positive correlation is found between STAT3and VEGF in salivary adenoid cystic carcinoma(P <0.05), between STAT3and bFGF in Salivary adenoid cystic carcinoma(P <0.05), between bFGF and VEGF in salivary adenoid cystic carcinoma(P <0.05).6The mean MVD is closely correlated with the expression of STAT3mRNA (P<0.05), and with the expression of VEGF mRNA (P <0.05), positive correlation is found between STAT3mRNA and VEGF mRNA in Salivary adenoid cystic carcinoma(r=0.390, P<0.05).In conclusion, STAT3、VEGF and bFGF positively correlate with MVD in salivary adenoid cystic carcinoma, which indicated that the expressions of STAT3、VEGF、bFGF may be responsible for the development of tumor and metastasis by promoting angiogenesis.The results show that there are positive correlation relationships among STAT3、VEGF and bFGF,which indicated that STAT3boost the development of new blood vessel by up-regulating the expression of VEGF and bFGF in the process of the development of the salivary adenoid cystic carcinoma. STAT3may become the new major treatment target of current antiangiogenic therapies in salivary adenoid cystic carcinoma.
