| Objective:The study was designed to study effect of absorbing components from Danggui-buxue tang on rat isolated thoracic aortas and the its possible mechanism.Methods:Absorbed components following oral Danggui-buxue tang were detected by UPLC-MS/MS method and used when alone or in combination to study the effects on tension response of isolated artery rings and the related mechanism.Results:1.The concentration of ferulic acid(m/z192.94>133.97), astragaloside â…£(m/z807.38>627.38), formononetin(m/z266.96>252) and calycosin(m/z282.97>268.03) in rat plasma after oral administration of Danggui-Buxue was123.10±3.21ã€54.32±1.24ã€17.41±0.36and14.34±0.35ng/ml respectively.2.The four components used alone or in combination all can concentration-dependently reduced the contraction(P<0.05). And there was significant different vasorelaxant between the rings with intact and denude endothelium in the first three(P<0.05).3.There was significant different vasorelaxant between incubated with L-NAME and without any blocker for the first three respectively(P<0.05).But calycosin doesn’t have this difference(P>0.05). 4.There was significant different vasorelaxant between incubated with (?)ndo and without any blocker for Astragaloside IV, formononetin respectively(P<0.05).But ferulic acid, and calycosin don’t have this difference(P>0.05).5.There was significant different vasorelaxant between incubated with Gliã€4-APã€BaCl2respectively and without any blocker for Astragaloside IV, calycosin(P<0.05). There was no significant different vasorelaxant between incubated with Gliã€BaCl2respectively and without any blocker for formononetin(P<0.05). But there was no significant difference between incubated with4-AP for formononetin(P>0.05). There was no significant difference between incubated with Gliã€4-APã€BaCl2respectively and without any blocker for ferulic acid(P>0.05).Conclusion:1. UPLC-MS/MS could determine absorbing components of Danggui-buxue fast, accurately, steadily.2. Absorbed compounds FAã€AS-â…£ã€FORã€CAL, they all have vasorelaxant on isolated rat artery rings;and when usd in combination induced optimal in-vitro vasorelaxant activity compared with them each.3. The relaxtion effect of FA is via endothelium-dependent mechanisms and relevant to the production of NO. While PGI2doesn’t participate FA’s relaxtion effect. KATP channel, Kv channel and KIR channel may be not involed either.4. The relaxtion effect of AS-IV is via endothelium-dependent mechanisms and relevant to the production of NO and PGI2. KATP channel, Kv channel and KIR channel may participate AS-IV’s relaxtion effect too.5. The relaxtion effect of FOR is via endothelium-dependent mechanisms and relevant to the production of NO and PGl2. KAtp channel and KIR channel may participate FOR’s relaxtion effect, while Kv channel may be not involed either.6. The relaxtion mechnism of CAL isn’t via endothelium-dependent mechanisms, isn’t relevant to the production of NO and PGL2. KATp channel, Kv channel and KIR channel participate CAL’s relaxtion effect. |
