"Reconciling Shaoyang":the Effect Of Xiaochaihu Tang And Electro Acupuncture At Foot Shaoyang On Osteoporosis Rats In Bone Mineral Density, Bone Histomorphology

objective:On the basis of the early stage research results of the subject "Shaoyang Meridians being in charge of the bone",——electro acupuncture at Foot Shaoyang Channel Points "adjust" the bone reconstruction,verify the effect of classical prescription Xiaochaihu Tang "Reconciling Shaoyang" on ovariectomy induced osteoporosis rats in bone mineral density, bone histomorphology, to make new realization of "Reconciling Shaoyang", in order to make the theory of traditional Chinese Medicine more perfect.Methods:Forty SD female rats aged6months according to completely randomized scheme were randomized into four groups of sham operation group, model group, gallbladder channel point group, and Xiaochaihu Tang group, ten in each group. Xiaochaihu Tang group were randomized into three groups, the rats number of middle dose group is4, both the middle dose group and low dose group is3(This stratification is another part of the subject, not included in this discussion).After found the Bilateral ovarian of sham operation group rats, remove the same size fat as the Bilateral ovarian. The other three groups were ovariectomized. Three months after operation, the gallbladder channel point group were treated with electro acupuncture at the corresponding points; Xiaochaihu Tang group were given with intervention of low dose, middle dose and high dose Xiaochaihu Tang intragastric administration; sham operation group and model group were not given any intervention. Three months after intervention the experiment finished, then the samples were collected for studying the indexs of bone mineral density(BMD), bone histomorphology.Results:1.Three months after intervention, in the model group, the BMD/body weight adjusted value were significantly lowered than those in the gallbladder channel point group, sham operation group and Xiaochaihu Tang group, the difference was statistically significant(P<0.01); The gallbladder meridian group (OVX-A) compared with sham operation group (Sham group) make no significant difference(P>0.05); Xiaochaihu Tang group compared with gallbladder meridian group and sham operation group also have no significant difference(P>0.05).2.Bone tissue HE stained observation showed that the bone microstructure in gallbladder channel point group and Xiaochaihu Tang group were more significantly improved than that of the other two groups.3.The number of osteoblast on the distal femoral metaphysis at the right of the rats:the number of osteoblast in model group were significantly decreased than those in the gallbladder channel point group, sham operation group and Xiaochaihu Tang group, the difference was statistically significant(P< 0.01); The gallbladder meridian group (OVX-A) compared with sham operation group (Sham group) make no significant difference(P>0.05); Xiaochaihu Tang group compared with gallbladder meridian group and sham operation group also have no significant difference(P>0.05).4.The number of osteoclast on the distal femoral metaphysis at the left of the rats: the number of osteoclast in model group were significantly increased than those in the gallbladder channel point group, sham operation group and Xiaochaihu Tang group, the difference was statistically significant(P<0.01); The gallbladder meridian group (OVX-A) compared with sham operation group (Sham group) make no significant difference(P>0.05); Xiaochaihu Tang group compared with gallbladder meridian group and sham operation group also have no significant difference(P>0.05).Conclusion:1.Classical prescription XiaochaihuTang"Reconciling Shaoyang" improve ovariectomy induced osteoporosis rats in bone mineral density and bone histomorphology, suggest XiaochaihuTang "ReconcilingShaoyang" has the effect of adjustment" on bone reconstruction, the effect of "regulating Yin and Yang". Expand the clinical use of "Reconciling Shaoyang".2.Both classical prescription of Xiaochaihu Tang "Reconciling Shaoyang" and electro acupuncture at Foot Shaoyang Channel Points has the effect of of "regulating Yin and Yang" on bone reconstruction, the mechanism of "adjustment" on bone reconstruction of XiaochaihuTang "Reconciling Shao yang" maybe as possible as the mechanism of electro acupuncture at Foot Shao yang Channel Points in ovariectomized induced osteoporosis rats of "adjustment" on bone reconstruction, This mechanism need to do further study from the level of molecular biology.
