Based On JAK2-STAT3 Signal Pathway Exploring The Effect Of Electroacupuncture On Shaoyang Meridian Points On Lipid Metabolism

Objective :In the previous experiments,it was confirmed that the electroacupuncture on Shaoyang meridian points had an effect on the lipid metabolism of normal SD rats[1].Based on this view,we want to observe the changes of serum leptin,blood glucose,blood lipid,insulin and hypothalamic leptin receptor(ob-R)and phosphorylated-signal transduction and activators of transcription3(P-stat3)in normal rats after electroacupuncture on Shaoyang meridian points.Through the peripheral system and the central nervous system(CNS),explore the reasons for how electroacupuncture on Shaoyang meridian points regulates lipid metabolism in normal rats,and how it can reduce the rats’ body fat,analyse the relationship between "bone" and "fat" on the theory of " Shaoyang in charge of bone ",and then provide more scientific experimental evidence for the prevention and treatment of obesity by using the electroacupuncture on Shaoyang meridian points.Methods:1、Grouping the animals: 60 SD rats,SPF grade,half male and half female,randomly divided them into female electroacupuncture on Shaoyang Meridian points group,male electroacupuncture on Shaoyang Meridian points group(referred to female EA group,male EA group),female control group,male control group,female blank group and male blank group,there were 6 groups with 10 rats in each group.2、Intervention methods: After the EA group rats were ligated,treated them with electroacupuncture on Shaoyang Meridian points,which were Huantiao point(GB30),Yanglingquan point(GB34)and Xuanzhong point(GB39).The control group rats were ligated as the same way as EA group,but did not accept EA treatment.Blank group rats did not receive any stimulation.3、Detection methods:3.1 General observation: Measured the body weight,the diet and the drinking water consumption of the rats at the same time every day,and observed their fur’s color,state of mind and defecation.3.2 Body fat observation: Separated and weighed the abdominal fat in rats,calculated the ratio of intra-abdominal fat / body weight;after wiping out the hair,the head and the viscera,calculated the rats’ subcutaneous fat and specific gravity by the formula.3.3 Serological indicators test: On the 5th,12 th,19th,and 26 th day of the experiment,cut the tail of all rats,collected about 1ml of blood from each rat,then separated the serum for detecting serum leptin.When the experiment was finished at the 26 th day,anesthetized the rats,taked out about 8ml of blood from the abdominal aorta,separated the serum for the detection of triglyceride(TG),cholesterol,blood glucose,and insulin.3.4 Detection of ob-R and P-stat3 in the hypothalamus:Opened the head of the rat,removed the intact brain tissue and placed it on the ice surface,quickly separated the hypothalamus on the ice surface,and immediately stored the hypothalamus in the-80℃ refrigerator.Used the method of Immunohistochemical frozen section to observe and compare the changes of ob-R and P-stat3 protein content in all the rats’ hypothalamus.Results:1、General situation: During this experiment,the rats in the EA group had shiny,supple fur,without abnormal behavior and were emotionally stable.During the time of receving the ligature and EA treatment they were gentle,and their stools were formed.The rats in the control group had slightly dull,messy fur,and their behavior were normal.However,when they were ligatured,the resistance was obvious,their emotion was violent,and their stools were not formed frequently.About the rats in the blank group,their stools were not formed occasionally and then they had no other abnormalities.2、Comparison of body weight,diet and drinking water consumption: Body weight: There was no significant difference in body weight between the same sex group(P>0.05).Diet: compared with the female blank group,the diet of the female control group was reduced(P<0.05),there was no significant difference in diet between any other two groups(P>0.05).Drinking water consumption: compared with the male blank group,the drinking water consumption in the male EA group was increased(P<0.05).There was no significant difference in drinking water consumption between any other two groups(P>0.05).3、Comparison of intra-abdominal fat,subcutaneous fat and specific gravity: intra-abdominal fat: compared with the control group and the blank group,the intra-abdominal fat in the EA group rats was significantly decreased(P<0.01);compared with the blank group,the intra-abdominal fat in the control group rats had no significant difference(P>0.05).Subcutaneous fat and specific gravity: female: compared with the blank group,the subcutaneous fat in the EA group rats was significantly decreased(P<0.01),and the specific gravity was significantly increased(P<0.01);compared with the control group,both subcutaneous fat and specific gravity in the EA group rats had no significant difference(P>0.05).Compared with the blank group,the subcutaneous fat in the control group rats was significantly decreased(P<0.01),and the specific gravity was significantly increased(P<0.01).Male: Compared with the control group and the blank group,the subcutaneous fat in the EA group rats was significantly decreased(P<0.01),and the specific gravity was significantly increased(P<0.01).Compared with the blank group,both subcutaneous fat and specific gravity in the control group rats had no significant difference(P>0.05).4、serum leptin concentration comparison:4.1Comparison between groups:After 4 weeks’ long experiment,compared with the blank group,serum leptin concentration in the control group rats had no significant difference(P>0.05);the concentration of serum leptin in the EA group was decreased(P<0.05).Compared with the control group,the concentration of serum leptin in the EA group rats was decreased(P<0.05).4.2Comparison within the group:EA group: There was no significant difference in serum leptin concentration between the 1th,2th,3th and 4th week(P>0.05).Control group: Compared with the 1th week,there was no significant difference in serum leptin concentration between the 2th and the 3th week(P>0.05),and the concentration of serum leptin in the 4th week was increased(P<0.05).Blank group: There was no significant difference in serum leptin concentration between the 1th,2th,3th and 4th week(P>0.05).5 、 blood glucose,TG,cholesterole comparison:5.1 blood glucose: Compared with the blank group,the blood glucose in the control group rats was increased(P<0.05),and the blood glucose in the EA group rats had no significant difference(P>0.05).Compared with the control group,the blood glucose in the EA group rats was reduced(P<0.05).5.2 TG: Compared with the blank group,TG in the control group rats had no significant difference(P>0.05),and TG in the the EA group rats was decreased(P<0.05).Compared with the control group,TG in the EA group rats was decreased(P<0.05).5.3 cholesterol: Compared with the blank group,cholesterol in the control group rats had no significant difference(P>0.05),and cholesterol in the EA group rats was increased(P<0.05).Compared with the control group,the cholesterol in the EA group rats was increased(P<0.05).6、Comparison of hypothalamic ob-R: Compared with the blank group,the hypothalamus ob-R protein content in the control group rats had no significant difference(P>0.05),and the hypothalamus ob-R protein content in the EA group rats was increased(P<0.05).Compared with the control group,the hypothalamus ob-R protein content in the EA group rats was increased(P<0.05).7、Comparison of hypothalamic P-stat3: Compared with the blank group,the hypothalamus P-stat3 protein content in the control group rats was increased(P<0.05),and the hypothalamus P-stat3 protein content in the EA group rats was increased(P<0.05).Compared with the control group,the hypothalamus P-stat3 protein content in the EA group rats was increased(P<0.05).Conclusions : 1 、 Electroacupuncture on Shaoyang Meridian points can affect the lipid metabolism of normal rats at 6 weeks’ age,and can reduce the body fat of normal rats,including subcutaneous fat or intra-abdominal fat.The weight,diet and water consumption of rats may be influenced.2、26 days’ long electroacupuncture on Shaoyang meridian point treatment,can reduce the concentration of serum leptin in normal rats,and then maintain its stability level,at the same time can increase the protein content of ob-R and P-stat3 in hypothalamus of normal rats.The cause of improving the lipid metabolism in normal rats after electroacupuncture may be related to the activation of JAK2-STAT3 signaling pathway.3 、Electroacupuncture on Shaoyang Meridian points can reduce TG in normal rats and may increase cholesterol in normal rats.4、Ligature may cause normal rats mood fluctuations,cause blood glucose rised,electroacupuncture on Shaoyang meridian points may can reduce blood glucose,so that it can return to normal level,at the same time,ligature may have a negative impact on normal rats’ emotional activities,electroacupuncture on Shaoyang Meridian points may regulate this negative effect,relieve the mood of rats,and improve the stool and the fur color abnormalities in rats.
