| OBJECTIVE:To investigate the anti-cancer effect of Si ni Decoction Treatment in hepatocellular carcinoma,and reveal the anti-cancer mechanism of the treatment in immune system and tumor immune microenvironment,in order to supplying theoretical and experimental evidence for warming Yang method in clinic.METHODS:Established the hepatoma model by injecting106Hepal-6cells into C57BL/6J mice’s liver. Observed the changes of tumorã€spleen index and thymus index. Flow cytometry (FCM) is used to detecte CD3+CD4+ã€CD3+CD8+ã€NKã€CD4+CD25+Foxp3+regulatory T cell subgroups in spleenã€bloodã€tumor and liver in order to analysis the effect of Si ni Decoction in immune system.RESULTS:(1) Si ni Decoction had a dose-dependent effect on inhibiteing the growth of tumor. High concentrate group of Si ni Decoction had an inhibition rate being more than30%and statistically significant (P<0.05).(2) Si ni Decoction could increase the Spleen index,but no obvious effect on thymus index.(3) Si ni Decoction could increase the proportion of CD3+CD4+and CD3+CD8+T cell in spleenã€blood and liver; increased the proportion of NK cell in spleen and liver; increased the ratio of CD4+/CD8+in blood and spleen,but no statistical significance in spleen; decreased the proportion of CD3+CD4+ã€CD3+CD8+and NK cell in tumor; decreased the proportion of CD4+CD25+Foxp3+regulatory T cell in spleenã€bloodã€tumor and liver.Among them, middle concentrate group had statistical significance(P<0.05).CONCLUSIONS:Si ni Decoction had an effect on inhibiteing the growth of tumor.Regulating the immune system and tumor immune microenvironment was one of the anti-cancer mechanism. |
