Purpose: This research project is a basic research,velvet antler polypeptides have avariety of biological effects. Deeplyreveals the mechanism of velvet antler polypeptides inhibits the chondrocyte apoptosis.Cofilin signaling pathway is an important signaling pathway, and its in-depth study isexpected to provide ideas to further clarify the pathogenesis of orthopedic clinicaldegenerative joint disease, as well as antler polypeptides on chondrocytes cofilin signalingpathway lay the theoretical foundation and provide the basis.Method: detect the normal rabbit cartilage cells by flow cytometry, cartilage cells wereimpacted after velvet antler polypeptides, rabbit chondrocytes of silent cofilin gene, thechanges of proliferation and apoptosis of hondrocytes of silent cofilin gene impacted aftervelvet antler polypeptides..Results: In this study, rabbit cartilage cycle by flow cytometry using Annexin V-FITCapoptosis detection kit early apoptotic cells, we found that the experimental group and thenormal control group, the experimental group the proportion of early apoptotic cells wassignificantly higher (P <0.001), this shows that the PAP can significantly reduce theproportion of early apoptotic cells, showed a good dose-effect relationship. Cartilage earlyapoptotic cells was significantly inhibited, has a protective effect on chondrocyte apoptosis.This provides a theoretical basis for PAP applied to the treatment of osteoarthritis of the drugafter. In the above experimental research foundation, further Cofilin gene silencing observedin the compressive stress stimulation rabbit cartilage cell morphology changes and apoptosis.And first by transfection psuper cofilin carrier, the cofilin gene silencing, given compressivestress irritation was observed after the rabbit cartilage cell morphology, found that the controlgroup: the cartilage cells of uniform size, the cell membrane is smooth, strong sense ofthree-dimensional cell refraction, the experimental group: chondrocytes unequal size, thegiant cells and some cells of irregular shape, cell spacing, refractive poor cell vacuoles. Asreorganization of cytoskeletal proteins in the signal path cofilin play in the maintenance ofcell morphology biological effect, but the exact mechanism remains to be further studied.Changes of rabbit chondrocyte apoptosis by flow cytometry, we found no significantdifference compared to the experimental group and control group, Although the compressivestress stimulation, cofilin signal path start, and play an important biological effects, but inrabbits it does not play a decisive role in the apoptosis of chondrocytes.
