Dendrobium huoshanense commonly known as Dendrobium mi, as the fresh or dried stems of orchids, is a precious medicinal herb in Anhui. Dendrobium huoshanense contains a variety of medicinal ingredients:Dendrobium polysaccharide dendrobine, protein and20kinds of Essential amino acids. Thus with the functions of eyesight, antitumor, antioxidant and hypoglycemic. At present, research on Dendrobium huoshanense focused on new techniques of cultivation and the chemical constituents and pharmacological effects. This paper focuses on the study of extraction and Purification of alkaloids and the process of vacuum freeze drying of Dendrobium huoshanense. Provide technical basis for the study of processing and alkaloids in the Dendrobium huoshanense. The main research contents are as follows:1.Effect of enzymatic hydrolysis conditions of extraction of alkaloids from Dendrobium huoshanense was studied. Discussion on the single factor of cellulase concentration, enzymolysis time, the effects of temperature and pH on extraction rate, then orthogonal test was tested. The optimum conditions of enzymatic hydrolysis:cellulose concentration is0.1%, hydrolysis time is2h, enzymolysis temperature is40℃, enzymatic is pH5.2.Alkaloid extraction of Dendrobium huoshanense can reach489.8μg/g.2.Study on the macroporous resin purification conditions of alkaloids in Dendrobium huoshanense..The comparison of adsorption of the AB-8, D101, NKA-9three resin for extract of Dendrobium huoshanense, the purification of AB-8macroporous adsorption resin is most suitable for Dendrobium huoshanense alkaloid. The effects of AB-8macroporous adsorption resin purification alkaloids of Dendrobium huoshanense process of eluant concentration, dosage, pH.Conclusions for Dendrobium huoshanense alkaloid purification parameters is:repeated sampling, the adsorption of pHlO.O, purified by elution with ethanol solution of agent70%and90%and the elution of pH4.0, elution volume is3BV.3.The extraction of total alkaloids of Dendrobium huoshanensewith Soxhlet extraction. Extract by extracting, macroporous resin purification, filtrating with membrane and the total alkaloid components of Dendrobium huoshanense were analyzed and identified by GC-MS method. GC-MS method detected12kinds of alkaloids, mainly for Dendrobium alkaloid, indole and pyridine alkaloid. There are two unknown alkaloid.GC-MS method to initial analysis of Dendrobium alkaloid, pyridine alkaloids quickly. The experimental results provide the basis for further development of alkaloids of Dendrobium huoshanense.4.Test the color of Dendrobium huoshanense. Discussion on the concentrations of acid zinc glucose,blanching time, blanching temperature and blanching effect of pH on Dendrobium huoshanense. The optimal process is as follows:the concentrations of acid zinc glucose was400mg/L, Blanching Temperature was90℃, blanching time was30s, pH8.0of blanching.5.The key process parameters affecting the quality of the products of vacuum freeze drying of Dendrobium huoshanense were studied. Freezing temperature was determined by measuring the eutectic point, and single factor test of the freezing time, heating temperature and vacuum were studied. The response surface design was on the basis of single factor test.A complex mathematical model was build between rehydration rate(Y) and each factor:Y=300+3.13X1+3.75X2+15.88X3-1.25X1X2+3.50X1X3-0.25X2X3-4.00X1X1-13.25X2X2-1.50X3X3, precise optimization of process parameters of vacuum freeze drying.Key of Dendrobium huoshanense freeze-drying process parameters:freezing temperature is-40℃, pre-freezing time is1.9h, heating temperature control in50.2℃, vacuum is75.0Pa.
