ObjectivesTo screen for the antifungal active substance from water-soluble extractfrom Asterias amurensis.And observe antifungal skin effect,reference to reducethe nurses hand fungi amount. Enhance the awareness of the medical staffhand hygiene, attention to nosocomial fungal infections, and improve the qualityof hand hygiene to reduce the incidence of nosocomial fungal infections.Methods1、Activity analysis experiment in vitro:The whole bodies of Asterias amuren-sis were chopped and extracted with water after remove the fat by organicsolvent.The extracts were separated by high performance liquid chromatograp-hy (HPLC) connecting the sephadex column in accordance with molecularweight.The all separated fractions of the water extracts were co-cultured withthe spore of fungus respectively. The spores concentrations of all groups werecounted to reflect the components of separation to inhibit fungals’ growthrespectively.2、 Activity analysis experiment in vivo: the fungi infection model of guinea pigskin were established and first separation fraction3(FSF-3) made intoointment to therapy and observe the influence to the skin lesions.3、The effect of the nurse hand hygiene were observed: Set the extractssolution group and conventional hand sanitizer to wash nurse’s hands andcompared with hands fungal colonies counts between two groups after washingdiscrepancy. Results1、The results of activity analysis experiment in vitro showed the first separatedfractions3(FSF-3) and FSF-4have the fungistasis to Tricophyton rubrum,Trichophyton tonsurans and Microsporum gypseum. Meanwhile that the obviousinhibition for the Tricophyton rubrum were observed by FSF-3. The inhibition ofTricophyton rubrum’ spore is dose dependent by FSF-3.2、The results of the animal experiments show that FSF-3can improve the skinlesions of fungal infections significantly. The FSF-3group have a significantdifference with no therapy group by score of treatment effects (p<0.01) and theFSF-3group have no significant difference with fluconazole therapy group(p>0.05).3、Observation on the effect of nurse hand hygiene showed that bothwater-soluble extracts solution and conventional sanitizer can decrease thefungal colonies (p<0.01).But the colonies count show anti-fungal skin effect ofwater-soluble extracts solution were equivalent to conventional sanitizer effectsafter wash hands (p>0.05).Conclusions1、The FSF-3have the fungistasis to Tricophyton rubrum,Trichophyton suransand Microsporum gypseum in vitro. The inhibition of Tricophyton rubrum’ sporewas dose dependent by FSF-3.The results suggested that the active substancethat inhibit the growth of Tricophyton rubrum were contained by FSF-3.2、The FSF-3separated from the water-soluble extracts of Asterias amurensisin SCS has significantly antifungal avtivity to Tricophyton rubrum on the guineapig skin fungi infection model, which suggests it may contain new antifungalactive substances.3、Observation on the effect of nurse hand hygiene: water extract solutioncontaining the active substance to improve the hand hygiene.
