Objective. To investigate the effects of sufentanil preconditioningon myocardial ischemia reperfusion injury in rats in vivo.Methods. To randomly divide60male SD rats equally into4groups,including:1. The Sham group(Group S), in which the chest wasopened without litigating the left coronary arteries (LADs);2. theIschemia-reperfusion Group(Group I/R), in which LADs are ligatedfor30minutes and are re-perfused for120minutes;3. The IschemicPreconditioning Group(Group IPC), in which the LADs are litigatedfor5mins and re-perfused for5mins with3replicates before30-mins-litigation of the LADs and120-mins-reperfusion;4. TheSufentanil Preconditioning Group (Group SUF), in which1.0μg/kg.Sufentanil is given by intravenous injection for5mins and rested for5mins with3replicates before30-mins-litigation of the LADs and120-mins-reperfusion (the total dose is3.0μg/kg).The following items are observed and calculated:1. to record theHR、MAP、LVDP and±dp/dtmax at different times in all groups;2.to measure the myocardial infarction size (IS/AAR%) withdouble-staining with Even’s blue and triphenyltetrazolium chloride;3.to calculate the concentration of cTnI;4. to calculate the concentrationof the serum malondialdehyde(MDA) with Thiobarbituric acid (TBA) reaction method and the activity of the superoxide dismutase (SOD)with xanthine oxidase reaction method;5. to observe myocardiummorphology by HE staning;6.to observed the apoptosis with TUNELmethod;7. to observe the expression of Bcl-2and Bax by theimmunohistochemical Staining and to observed the OD of Bcl-2andBax by semi-quantitative analysis.Result.1. Comparing with that in Group1/R, the cardinal functionsin Group IPC and Group SUF are enhanced according to the value ofthe LVDP and±dp/dtmax in all groups.2. Comparing with that inGroup1/R, the myocardial infarction size(IS/AAR%) in Group IPCand Group SUF decreased obviously.3. Comparing with that inGroup1/R, the concentration of cTnI in Group IPC and Group SUFdecreased obviously;4. Comparing with that in Group1/R, theconcentration of the serum malondialdehyde (MDA) in Group IPCand Group SUF reduced and the activity of the superoxide dismutaseincreased.5. Comparing with that in Group1/R, the cellmorphological observation in Group IPC and Group SUF show lesschanges in pathology.6. Comparing with that in Group1/R, thequantity of the cardiac apoptosis in Group IPC and SUF reducedobviously.7. Comparing with that in Group1/R, the OD value ofBcl-2gens and the value of Bcl-2/Bax increased, and the expressionof Bax gens decreased. Conclusion. Sufentanil preconditioning has protective effects onmyocardial ischemia-reperfusion injury in rats in vivo.
