| Valeriana jatamansi Jones is a common Chinese medicine. Based on the review on the chemical constituents and pharmacological researeh of Valeriana jatamansi Jones, the chemical constituents of the alcohol extract were studied and their antitumor activities were tested.With cold-maceration, dry powder of Valeriana jatamansi Jones was extracted four times by95%ethanol, each time24h. Extract was prepared with vacuum concentration. Then the extract was extracted by petroleum ether, ethyl acetate, n-butanol, to divide into three parts. The ethyl acetate extract were separated preliminary with silica gel chromatographic methods. Then, various chromatographic methods such as silica gel, Sephadex LH-20, RP-C18column chromatography was used over and over again to separate and purify the compounds. Finally, Thirteen compounds have been isolated.The structures of the thirteen compounds which had been identified were elucidated by EI-MS,1H-NMR,13C-NMR, COSY, HSQC, HMBC, and compared with the known compounds. They were identified as:Daucosterol (1), Oleanic acid (2),(β-Sitosterol (3), Valeriananoid C(4), Valjatrate D (1,7-isovaleryloxy-8-methyl-3,8-epoxyvalechlorine)(5), Chlorovaltrate A (6),4-Hydroxy-3-methoxycinnamaldehyde(7), Didrovaltratum-valerox-yhydrin (8),1-methoxy-7-hydroxy-8-methyl-3-epoxy-Δ4,11-dihyronepetane(9), Jatamanvaltrate G (10), Valtratum-isovaleroxyhydrin(11), Valjatrate E(12)ã€Rupesin E(13). Among the total, three sterides including compound1-3, eight iridoids including5-6,8-13; Compound4is a sesquiterpenes, and Compound7is a cinnamaldehyde. Compound5and12are identified as new structures by searching data-base of SciFinder Scholar. Compound7were isolated from the plant for the first time.The compound4,6,10,11,12,13was be tested the in vitro cytotoxicity activities against MCF-7cancer cell lines. The effeets of compounds on human cancer cells growth, expressed as the IC50value, for MCF-7cells lines compounds showed growth inhibitory effects and differentiation induction abilities, compound12was the most potent with the IC50value of20nM. |
