| Ovarian cancer (Epithelial Ovarian Carcinoma, EOC) is a gynecologic malignant tumorof the highest mortality rate. Because of its occult onset, rapid progress,70%~80%patientswho were diagnosed have been advanced, which brings much difficulty to the clinicaltreatment. Although people continue to carry out the ovarian cancer research, the origin andpathogenesis are not fully clarified yet. In malignant ovarian tumor, epithelial ovarian cancer(Epithelialov arianc ancer, EOC) accounted for about90%[1-2]. Research has shown that,epithelial ovarian cancer is not an independent disease, but a different set of tumor, whichcan be classified according to characteristics of morphology and genetic molecular[3]. Atpresent, the standard therapy recommend by the International Federation of Gynecology andObstetrics (FIGO), and the National Comprehensive Cancer Network (NCCN) ischemotherapy, which is the cytoreductive surgery combined with paclitaxel and platinum,but due to the long period of chemotherapy, the tumor cells develop resistance to the drugplatinum and recurrence[4,5]. At the same time, the side effects and the expensive price ofchemotherapy drugs caused the therapy interruption and influenced the effect of treatment,which made the survival rate of the five year in patients with ovarian cancer have beenhovering at around40%[5]and most patients survived without tumor who reoccurred after18months[6]. Therefore, reducing the side effects of chemotherapy drugs and reversing ofovarian cancer cell drug resistance are the hot and difficult problems. And finding theinhibition of tumor growth, metastasis and reversal of drug resistance of pure naturaleffective preparation is very important.Chinese traditional medicine is Chinese nation’s essence. Chinese medicine in thetreatment of tumor has a long history in our country, which has been attracted internationalattentions in recent years, because of good anti-tumor effect, small side effect and low costadvantages of traditional Chinese medicine. The roots of Anemone raddeana Regel (A.raddeana Regel), in traditional Chinese medicine is called pointed at both ends, and alsoknown as anemone raddeana, are traditional Chinese medicine and have the functions of rheumatism and removal of swelling, which can be used for treating wind cold dampnessarthralgia, joint pain, limb spasm, carbuncle swollen canker disease[7]. With the developmentof unceasing study of modern pharmacology, people found the anemone raddeana Regel inswelling analgesic, antibacterial, immune regulation and anti tumor has good pharmaceuticalvalue.In1980s, anemone Ais separated from the drug with anticancer activity[8], and withoutthe side effect of Mutation of chromosome[9]. The extractions of Anemone raddeana Regelhave better antitumor effect in vitro and in vivo. Inducing tumor cell apoptosis is animportant way to play its anti tumor effect, but also can play the anti tumor effects throughthe mechanism of immune regulation[10]. Scholars have found, raddeanin A has significantinhibitory effect on colon cancer, lung cancer, liver cancer and cervical cancer, but studies inovarian cancer is rare.This paper uses the extraction of anemone raddeana Regel raddeanin A as the researchobjective to explore the effects of different concentrations of Anemone A monomer on Skov3ovarian cancer cell proliferation, apoptosis and invasion; detect the genes transcription ofBax, Caspase3, Bcl-2, Stat3, MMP-2, MMP-9, Cyclin D1by Real time PCR and theprotein expression of Cyclin D1, Cyclin B1, Bcl-2, Cyt-c, Caspase-3, Bcl-xL, Bax, MMP-9,MMP-2, Stat3by Western-blot; explore its mechanism to provide a theoretical basis for thetreatment of ovarian cancer and its clinical application and further development. |
