Objective:To study the influence of traditional Chinese herb qianzhi capsule and baizhi on the cell growth factor secretion and the related gene expression of human endometrial stromal cells and glandular epithelial cells in vitro.Methods:The human endometrial tissue was cultured in vitro. Part of the cells were cultured on cover glasses to identify the cell purity with specific antibodies, the others were incubated with different concentrations of qianzhi capsule and baizhi for24h,48h and72h to determine its cell proliferation activity on the endometrial glandular epithelial cells and stromal cells by MTT method. The cell culture were harvested after treatment of qianzhi capsule and baizhi for48h, then the ELISA method was used to determine the expression of cytokines. Gene expressions of endometrial stromal cells and glandular epithelial cells treated with qianzhi capsule and baizhi were detected by RT-PCR. The Ca2+content in endometrial glandular epithelial cells treated with qianzhi capsule and baizhi were also detected by the fluorescent labeling method.Results:Chinese herb qianzhi capsule can promote the growth of endometrial glandular epithelial cells in the concentration of10-6mg/ml after incubation of48h. But Chinese herb baizhi did not have this effect.Both qianzhi capsule and baizhi can induce the mRNA expression of calcitonin (CT) of endometrial glandular epithelial cells and significantly increase Ca2+content of endometrial glandular epithelial cells. Meanwhile, it had no significant effects on the secretion of cytokines.Conclusion:Chinese herb qianzhi capsule can promote the growth of endometrial glandular epithelial cell in vitro, but not Chinese herb baizhi.However,both qianzhi and baizhi can induce the mRNA expression of calcitonin (CT) and significantly increase the Ca2+content of endometrial glandular epithelial cells. These results imply that Chinese herb qianzhi and baizhi could improve the receptivity of endometrium, which is beneficial to increase embryo implantation and clinical pregnancy rate. Meanwhile, the results indirectly reflected the safety of the Chinese herb qianzhi as it doesn’t have obvious effect on the secretion of cytokines, the results indirectly reflects the safety of the Chinese native medicine because of qian zhi have no obvious effect on the secretion of cytokines.
