Objective: To investigate the expression of endometrial COX-2at the timeof the implantation window in the women undergoing repeated implantationfailure following IVF-ET,and to explore the relationship between COX-2andendometrial receptivity. This study may provide a theoretical basis in order toimprove endometrial receptivity and implantation rate.Methods: A total of44Infertile women participated in this study anddivided into three groups. group A:18patients who failed of three cycles ormore following ET/FET;group B:15patients failed in the first cycle ET;groupC:13patients were pregnant in the first cycle ET.The mRNA expression ofendometrial COX-2at the time of the implantation window in44cases wasmeasured by Real-Time PCR.Results:1.To compare three groups to the women age, infertility fixednumber of year,BMI,type of infertility, infertility factor, fertilization method.Allof the index were not statistically significant,(P>0.05).2.To compare three groups to basic FSH value, the dose of gonadotropinand duration of stimulation, the E2and Prog level on theday of HCG injection, average number of retrieved oocytes, endometrial thickness on the day of ETadministration day.All of the index were not statistically significant,(P>0.05).3. The expression of endometrial COX-2mRNA in the three groups were0.0263±0.0257,0.0522±0.0309,0.0598±0.0326respectively,the differences werestatistically significant,(P<0.05). the COX-2mRNA expression in group Awas less than in group B and C,(P<0.05).The ratio of the difference betweentgroups C and groups A was2.3times. The ratio of the difference betweentgroups B and groups A was2times. No significant differeces were foundbetween groups B and groups C,(P>0.05).Conclusions:The expression of endometrial COX-2mRNA appears to bedecreased in patients with repeated implantation failure compared withsuccessfully pregnant in the first ET controls. We therefore suggest that thereduction of COX-2in the human endometrium at the time of theimplantation window may lead to poor endometrial receptivity.
