| Objective The paper aims to research how Didang Tang and its partial recipe to such method have the effect on treating diabetes. The research adopts to observe myocardial fibrosis and TGF-β1/Smads signal pathway of lab rats influenced by Didang Tang and its partial recipeMethods The research needs120Sprague Dawley rats and each rat weighs200-220g. After10hours that the120Sprague Dawley rats were food deprived but water given,105Sprague Dawley rats were intraperitoneally injected STZ in a dose of55mg/kg. After72hours STZ injection, the rats were measured glucose by caudal vein blood drawing. The rats whose glucose level>16.7mmol/L were regarded as successful DM module copies.100successful DM module copies were divided into5groups in random and each group had20rats.(1) Model group:Rats in Model group were given intragastric administration in1.0ml/100g distilled water every day.(2) Chinese medicine soup named Didang group:Every milliliter of distilled water was mixed with1g herbal Chinese medicine named Didang recipe. Rats in Chinese medicine soup named Didang group were given intragastric administration in1.0ml/100g of the mixed liquid.(3) Leech and gadfly group:Every milliliter of distilled water was mixed with1g herbal Chinese medicine named leech and gadfly recipe. Rats in leech and gadfly recipe group were given intragastric administration in1.0ml/100g of the mixed liquid.(4) Peach seeds and rheum officinale group:Every milliliter of distilled water was mixed with1g herbal Chinese medicine named Peach seeds and rheum officinale recipe. Rats in peach seeds and rheum officinale group were given intragastric administration in1.0ml/100g of the mixed liquid.(5) Valsartan capsule group:Rats in valsartan capsule group were given intragastric administration in80mg/kg of mixture with valsartan capsule and distilled water. No-treatment control group consisted of15normal rats who were given intragastric administration in1.0ml/100g of distilled water. All the rats in above six groups were continually given intragastric administration once a day for8weeks. At the end of the8th week, all the rats in above six groups were anesthetized and killed. We observed these rats’fasting blood glucose, HE chromosome and Masson chromosome. We adopted western blot to detect TGF-β1ã€Smad2ã€Smad3ã€Smad7in rats’myocardium tissue and adopted radio immunoassay to measure Angâ…¡ content in myocardium tissue.Resultsâ‘ Compared with no-treatment control group, fasting blood glucose of rats in model group was significantly up (p<0.01). Compared with model group, fasting blood glucose of rats in valsartan capsule group was significantly down. Fasting blood glucose of rats in Didang Tang and that of in decomposed Didang recipe did not have obvious change (p>0.05).â‘¡According to HE chromosome indication, myocardial cells of rats in no-treatment control group arranged in neat rows, cell size uniformity, homogenate staining. While myocardial fibers of rats in model group dissolved, arranged in a mess and were infiltrated by cells with inflammation. Myocardial cells of rats in Didang Tang group arranged in neat rows. Myocardial cells of rats in valsartan capsule group arranged in neat rows while a few myocardial fibers had breakage. Rats in leech and gadfly group and in peach seeds and rheum officinale group were myocardial fibers hypertrophy and myocardial fibers breakage.â‘¢According to Masson chromosome indication, quantity of collagens of rats in no-treatment control group was a few while arranged in a neat rows. Myocardial cells of rats in model group was a few, hypertrophy and breakage, which indicates collagen hyperplasia. Myocardial fibers of rats in Didang Tang group arranged in a neat row and did not have collagen hyperplasia. Compared with that of model group, myocardial fibers of rats in leech and gadfly group and in peach seeds and rheum officinale group had more breakage and collagen hyperplasia. myocardial fibers of rats in valsartan capsule group arranged in neat rows and had a few of collagen hyperplasia.â‘£Compared with no-treatment control group, contents of TGF-β1ã€Smad2ã€Smad3in diabetic cardiomyopathy model group were up (p<0.01) while contents of Smad7were down (p<0.01). In comparison with model group, contents of TGF-β1in the whole Didang recipe group and in its partial recipe as well as in valsartan capsule group were obviously down. Contents of smad2in Didang Tang group group, in peach seeds and rheum officinale group and in valsartan capsule group were significant down (p<0.01). While that in leech and gadfly group did not obvious change (p>0.05). Contents of Smad3in Didang Tang group and in its partial recipe group, in valsartan capsule group were significant down (p<0.01). Contents of Smad7in Didang Tang group, valsartan capsule group were significant up (p<0.01). Contents of Smad7in peach seeds and rheum officinale group were up (p<0.05) while that of in leech and gadfly group had no obvious change (p>0.05). In comparison with valsartan capsule group, contents of TGF-β1ã€Smad2ã€Smad3in leech and gadfly group were up (p<0.01) while contents of Smad7were down (p<0.05). contents of TGF-β1(p<0.05)ã€Smad2(p<0.01)〠Smad3(p<0.01) in peach seeds and rheum officinale group did have significant difference while contents of Smad7did not have significant difference (p>0.05) contents of TGF-β1ã€Smad2ã€Smad3in Didang Tang group did not have statistical meaning (p>0.05), Contents of Smad7in Didang Tang group were up (p<0.05). Compared with Didang Tang group, TGF-β1ã€Smad2ã€Smad3in leech and gadfly group and in peach seeds and rheum officinale group both had significant change (p <0.01). Referring to Smad7, Smad7in leech and gadfly group (p<0.01) and Smad7in peach seeds and rheum officinale group (p<0.05) both had significant change.⑤Compared with that in no-treatment control group, Ang â…¡ of myocardial fibers in model group was significant up(p<0.01) while Ang â…¡ of myocardial fibers in valsartan capsule group, in Didang Tang group and its partial recipe group were obvious lower than that in model group (p<0.01). In comparison with valsartan capsule group,Ang â…¡ of myocardial fibers in leech and gadfly group and in peach seeds and rheum officinale group was (p<0.05), Ang â…¡ of myocardial fibers in Didang Tang group and in valsartan capsule group did not have significant difference (p>0.05). Compared with Ang â…¡ of myocardial fibers in Didang Tang group, Ang â…¡ of myocardial fibers in leech and gadfly group and in peach seeds and rheum officinale group both had significant difference (p<0.05)Conclusion Didang Tang group has good effect on suppressing myocardial fibrosis of rats of DM models, which is similar to valsartan capsule group. Such good effect has relation to suppressing myocardial fibrosis, reducing Angâ…¡ content and influencing TGF-β1/Smads signal pathway caused by Angâ…¡. The effect of the whole recipe is better than that of the partial recipe. |
