Objectives To research the effect of the mouse serum which contain the Radix Echinopsis on the alpha microtubule protein acetylation level and molecular partner HSP90function of the human multiple myeloma cell line U266.Methods To detect the content of the alpha sanlian thiophene in the coarse extraction drugs by high-pressure liquid chromatography(HPLC); The acetylation level of a-tublin which was a substrate of HDAC6was detected by Western blot; Detecting the chemical force between the HSP90and its Client proteins by immune precipitation (IP).Results After the U266cells effected by the different concentrations of serum medicated (0,1,2,4g/ml)24hours or48hours, Western blot analysis showed that its lpha microtubule protein acetylation level and degree of acetylation of the HSP90gradually increased, and which changes in a dose dependent (P<0.05).Conclusion Radix Echinopsis had a concentration-and time-dependent effect on the Acetylation of the acetylated a-tublin and HSP90, which can induce the apoptosis of human multiple myeloma cell line U266and can be used as a new source of plant drugs in the treatment of multiple myeloma.
