| ObjectiveTo observe the effects of Yishentiaodu acupuncture-moxibustion on theAD(Alzheimer’s disease) rats with protein COX Ⅳabout energy metabolismof hippocampus mitochondria,and Cyp-D about permeability transition poreof hippocampus mitochondria. To investigate the specific mechanism ofprotect mitochondrial function and inhibition mitochondrial injury on AD byacupuncture-moxibustion. Further defined the specific mechanism ofprevention-treatment on AD by acupuncture-moxibustion, offering moretheoretical foundation and significant data to clinical care on AD.Methods40Wistar rats were selected through the Morris-maze screening (rejectedthe rats that reaction were too slow and too quick). According to randomnumber table,the qualified were randomly divided into4groups: the normalgroup, sham-operated group, model group, and acupuncture-moxibustiongroup. First, bilateral hippocampus were injected Aβ1-42to make rat model ofAD. Then, acupuncture-moxibustion group selected Baihui points andShenshu points, and treated with acupuncture and moxibustion. Last, detectthe expression of hippocampus mitochondria Cyp-D and COXIV in rats by themethod of immunohistochemistry. Analyzing the differences among thegroups through statistical. Results1.Immunohistochemistry result of Cyp-D protein showed that compared themean optical density and average gray with normal group and sham-operatedgroup, Cyp-D expression of model group’ hippocampus mitochondria ishigher obviously than that of them (P<0.01). However, compared with themodel group, Cyp-D expression of acupuncture-moxibustion group’shippocampus mitochondria decreased significantly (P<0.01).2.Immunohistochemistry result of COXIV protein showed that compared themean optical density and average gray with normal group and sham-operatedgroup, COXⅣ expression of model group’s hippocampus mitochondria islower obviously than that of them(P<0.01). However, compared with themodel group, COXⅣ expression of acupuncture-moxibustion group’hippocampus mitochondria increased significantly (P<0.01).Conclusions1.Yishentiaodu acupuncture-moxibustion fully meet the pathologic feature ofAD that have old age and efficiency of the kidney and encephalon and uselessof mysterious mechanism, which can effectively prevent the occurrence anddevelopment of AD.2.The Cyp-D expression of Yishentiaodu acupuncture-moxibustion caneffectively inhibit the Cyp-D expression of hippocampus mitochondria,blocking the formation of mPTP, and decreasing mitochondrial damaged.Thereby Yishentiaodu acupuncture-moxibustion plays a role in treatment andprevention for Alzheimer’s disease.3.Yishentiaodu acupuncture-moxibustion can increase the activity ofhippocampus mitochondrial respiratory chain enzyme complexes and ATPconcentration improve mitochondrial function. Thereby Yishentiaoduacupuncture-moxibustion plays a role in treatment and prevention for Alzheimer’s disease.4.The mechanism of prevent AD is that Yishentiaoduacupuncture-moxibustion might reduce mitochondria membrane permeability,improve the energy metabolism and break down the rate of the mitochondrialinjury, then prevent neurotoxicity from Aβ, maintain the viability of neuronsand play a protection part in neurons. Thereby Yishentiaoduacupuncture-moxibustion plays a role in treatment and prevention forAlzheimer’s disease. |
