Objective:It had been confirmed that the β-amyloid is the most important substance in the course of forming the senile plaque(SP) extracellular. And as we know, the senile plaque and the neurofibrillary tangles (NFT) have been testified as the most Characteristic pathological changes in the brain of the patients with Alzheimer’s disease. In view of that, there are two ways to cut down the (3-amyloid so that it could form less SP. One of the ways is to reduce the excess production of the β-amyloid, and the other way is to boost the degradation of the β-amyloid. Both the two methods are effective to refine the Alzheimer’s Disease’s Characteristic pathological changes, and to some extent to delay the development of the Alzheimer’s Disease. According to the literature which are available in the relative document reference at present, there have been a lot of articles in which the production process of the β-amyloid were studied with all kinds of techniques, and to some extent, the scholars had got some meaningful results from the studies. Therefore the respect of the β-amyloid’s production had been studied deeply. While, as to the process of the β-amyloid’s degradation, in spite of that there had been some studies which are available in some published magazines, compared with the process of production of β-amyloid, more and deeper researches are still needed to be done in the term of its degradation. In the light of the researches before, we had learned that there are the autolysosome system and the ubiquition pro tea some system, which are the main ways to degrade the β-amyloid in the cell. However, the two systems could not clear away the β-amyloid which is located outside the cell, after all the extracellular β-amyloid is the key to form the SP. As to the extracellular, it had been found that some proteases which had been experimented in vitro tests before could degrade the A β, such as Neprilysin, Presequence Protease, angiotensin converting enzyme, Fibrinolytic enzyme, matrix metalloproteinase-9and so on. In this research, we would pay our attention to the respect of the protein degradation of the β-amyloid. The Neprilysin and Presequence Protease, which are the proteases to degrade the Aβ, were chosen as the experiment indexes, we want to study the possible therapeutic mechanism on the Alzheimer’s disease with the treatment of acupuncture and moxibusition. With the usage of the immune-o-hist-o-chemistry technology, we would observe the expressing of the Neprilysin and Presequence Protease in the CA1part in rat’s hippocampus of four groups. And in this study we use the therapy of tonifiering the kidney and strengthening marrow with acupuncture and moxibusition to treat the AD rats.Methods:We chose the Wistar male rats as the object of this study, which were about4months old. After the examination of the Morris Water Maze, the eligible Wistar rats which were not too clever and not too fool, according to the random number table, were arranged into four groups:the model group, the sham-operation group, the treatment group and the normal group. The A β1-42in condensed state was injected into the bilateral hippocampus of the rats to prepare the AD rat model. As to the sham-operation group, the same quantity of normal saline would been injected into the bilateral hippocampus of rats in the same way as the model group. In the treatment group, the points of Baihui (GV20) and bilateral Shenshu (BL23) were selected for the treatment of acupuncture and moxibustion. As to the treatment, the needles (1inch28filiform needle) were first penetrated into the three points, After the acupuncture treatment, the thin moxa sticks (about5mm in diam) were lighted to be hanged above the three points with2-3centimeters high, and this warming moxibusition would also last15minutes. During the course of moxibusition. This treatment would be performed in the morning every day.6days as a course and there was a day to have a rest between courses. This research would last two courses. When the rats in the treatment group were treating, the rats in other three groups got the same condition to keep without any special processing. When the treatment came to an end, we would chose10rats in each group to test index. The immune-o-hist-o-chemistry was adopted to observe the expression of the Neprilysin and Presequence Protease in the CA1part in rat’ s hippocampus of four groups,Results:After the treatment of two courses, with the examination of The immune-o-hist-o-chemistry,we could find:(1) compared with the model group, the expressing of the NEP in the hippocampus of the rats was apparently lower in the normal group, indicating statistical significant difference (P<0.01), while the content of NEP in treatment group was higher significantly than those in model group(P<0.01);(2) as to the PreP, in the normal group, the quantity was more than in the model group(P<0.01); compared with the model group, the level of the PreP were much higher in treatment group(P<0.01), and the difference was remaekble.Conclusion:Under the treatment of acupuncture and moxibustion for modifying the Governor Meridian and tonifying the kidney, the quantities of the Neprilysin and Presequence Protease were increasing greatly. For the sake of that, it could help to degrade more the β-amyloid, therefore the aggregation and deposition of Aβ could reduce, so that it could delay the development of the Alzheimer’s disease to some extent. |
