| Objective:By observing the influence of Qidanhuatan Decoction on myocardial enzyme series.Vascular endothelial factor levels and the missing area cardiac troponin(cTnT) of acute myocardial ischemia in rat, to discuss it’s experimental mechanism of protective effect of ischemic myocardial injury in rats.Methods:Acute myocardial ischemic model was established by Intraperitoneal injection of pituitrin in rats, to determine the Serum NO.ET activity and content, to determine the average cTnT dyeing defect area by immunohistochemistry.60adult SD rats((half male and female) which are similar in weight and open-field test grade, were randomly divided into a blank group, model group. Qidanhuatan Decoction of low, medium and high groups, Enalapril Maleate groups,10in each group. The rat of Qidanhuatan Decoction of low, medium and high groups were given gavage respectively according to the corresponding concentration of1mL/100g dose, while the Enalapril Maleate groups were given gavage0.015g/kg according to the specification, the-blank group and model group were filled with0.9%saline solution volume, group, each of group was given dispose for7days.lh after the last administration, except the blank group, to give the rest of the rats by intraperitoneal injection of pituitary vasopressin (6U/kg),5sec after injection. Then anesthetized by intraperitoneal injection with10%chloral hydrate (3mL/kg), Sit-fixed.and by Picking up BL-420E+biological function experiment system.to recorde ECG lead standards.Record-lead standard ECG, adjustment the sensitivity1mV=15mm, ST-segment elevation0.1mv was modeling successful standard. On the record after the last ECG open the abdominal cavity, each rat abdominal incision after1hours, Abdominal aortic blood, low temperature separation serum for measurement. Each rat abdominal aortic blood of5ml, let stand for2hours, centrifugal (4000r/min) for10min, placed in the refrigerator-20under to test, CK, LDH. NO and ET content were measured according to kit instructions, to determine the average cTnT dyeing defect area by immunohistochemistry. Data processing with the statistical software spss12.0analysis.Results:(1) The influence of Qidanhuatan Decoction on ECG ST segment myocardial ischemia of rats caused by Pit, the model group rats Ⅱ lead ECG changes of ST segment was obviously higher than that of normal group,S-T segment elevation>0.1mv,which show the myocardial ischemia model success;(2) The influence of Qidanhuatan Decoction on serum myocardial enzyme, the serum of CK, LDH,ET content was significantly lower than that of normal group, while the NO was significant increase than that of normal group;(4) Qidanhuatan Decoction could reduce the lack of space of cTnT in early time; Qidanhuatan Decoction can improve the cardiac protection protective in acute myocardial ischemia, cardiac, which relate to reduce serum LDH and CK ET content, increase serum NO level, reduce the lack of cTnT area, it provide feasible basis for clinical treatment.Conclusions:(1) Qidanhuatan Decoction can obviously improve the acute myocar-dial ischemia rat ECG ST segment changes, alleviate myocardial ischemia effectively, has protective effect on acute myocardial ischemia.(2) Qidanhuatan Decoction may improve myocardial ischemia mechanism such as:reduce the Myoca rdial enzyme LDH activity of CK; raise NO level and reduce the ET Level, correct imbalance of NO/ET.(3) cTnT has different level of loss in early acute myocardial ischemia, and the lack of cTnT is not just within the cytoplasm is missing, which was characterized by muscle fiber protein structure collapse.Qidanhuatan Decoction by improving myocardial oxygen supply and reduce the degree and range of myocardial ischemia damage, protection of, to protect endothelial function and stabilize cardiac muscle cells, so as to achieve for the treatment of myocardial ischemia... |
