| Chlorophyll a is the main plant photosynthetic pigment, and its exist in most of Marine phytoplankton. In the view of the present increasingly serious Marine environment pollution, the chlorophyll a can real-time, accurate and sensitive assay for Marine red tide, green ecological disasters and so on to work for risk early warning. In addition, chlorophyll a has a close relationship with Marine fisheries and aquaculture. Which is one of the important testing project in Marine ecology.Many kinds of methods can detect the Marine phytoplankton concentration. Traditional chlorophyll test by the method of extraction have the defect that the test program consume much time and can not be used for continuous monitoring. So we are unable to get occur, early and real-time data in the first phase. With the Fluorescent characteristics of sensitive, rapid and accurate in the application of seawater, chlorophyll detection has become one of hotspots in the field of Marine monitoring. Find the rapid detection device of chlorophyll fluorescence to achieve the real-time test can provide a scientific basis for government decision-making of the ocean disaster early warning and accident like red tides, etc.This article summarizes various methods of determining of chlorophyll a and the advantages and disadvantages of each method are analyzed and compared.In comparison, fluorescence spectrometry is an efficient and sensitive method, which is suitable for field testing and online monitoring. However, the determination of chlorophyll a in water is easily affected by various environmental factors,resulting in the decline in the accuracy of the measurement data. This paper mainly studies the impact of temperature and turbidity on the determination of chlorophyll a with fluorescence method, explores the influence mechanism and extent. The results show that the temperature rise will make the fluorescence intensity of chlorophyll a drops and there’s a good linear relationship between them, R2 = 0.99; turbidity can also make the fluorescence intensity of chlorophyll a drops, there’s also a certain linear relationship, R2 = 0.98. And verify the affect of the temperature and turbidity on the determination of chlorophyll a in water by the on-line detector for seawater’chlorophyll respectively.Overall, the fluorometer is very stable, the relative error between determination value of the temperature experiments and true value within ±20% and the relative error between determination value of the turbidity experiments and true value within 5%, the effect of the two factors on the instrument is within the acceptable range. |
