Eucommia Ulmoides leaves contain a variety of medicinal components which are widely used in food, medicine and other fields. The leaching of chlorogenic acid(CAG) and flavonoids were studied in this work. The adsorption and desorption of the chlorogenic acid and the preparation of microcapsule were also researched.Ethanol was employed as a solvent for leaching CAG from Eucommia Ulmoides leaves and HPLC was used to determine the concentration of CAG in the extracts. The process variables, such as the ratio of liquid to solid, the ethanol concentration(φ), the leaching temperature and leaching time, and the leaching kinetics were investigated. The optimum parameters were summed as follows: liquid to solid ratio of 10 mL/g, ethanol concentration of 60%, leaching temperature of 60 ℃ and leachnig time of 40 minutes. Under the optimum conditions, the leaching yield of CAG reached up to 90.21%. The leaching process is kinetically controlled by internal diffusion, and the apparent activation energy is 11.46 kJ/mol.The ratio of liquid to solid, the ethanol concentration, the leaching temperature and the leaching time for flavonoids leaching. the optimum conditions are: liquid to solid ratio of 14 mL/g, φ= 72%, leachnig temperature of 84 ℃ and leachnig time of 2.4 h. The maximum yields of flavonoids is 23.12 mg/g. The microscopical preparation were also investigated. The results were as follows: the mixture of 1.5 g acacia and 1g β-cyclodextrin was found better to make the microcapsule, and the ratio of core material to wall material was 0.10 g/g.Macroporous resin, NKA-2, NKA-9, HPD600, HPD400, HPD100, D101, were compared for the purification and desorption of CAG. Effects of the pH value, sample concentration, and temperature on static adsorption of macroporous resin and the adsorption kinetics were investigated. NKA-2 was better for the purification and desorption of CAG. The pH value for CAG adsorption was 3, and the adsorption capacity rises while temperature drops, and the maximum adsorption amount was 70.15 mg/g at room temperature(25 ℃). The optimum ethanol concentration for the desorption was 50%. The optimum parameters for dynamic CAG adsorption are as follows: sample concentration of 1.0 mg/mL, flow rate of 2 mL / min. The purity of CAG can reach to 39.51%.
