Eucommia ulmoides Oliv is a very ancient tree species,about 200 million years ago,the invasion of the Quaternary glaciers causing the Eucommia plant extinction in Europe,Asia and North America.Due to the complexity of the topography in Central China,the glaciers are blocked.A kind of Eucommia ulmoides Oliv survived,and became China’s unique plant species.Eucommia ulmoide s Oliv is called ―pioneer plants‖ by scientists,which has a very tenacious vitality.Eucommia ulmoides Oliv is a unique Chinese herbal medicine.It has been found that Eucommia ulmoides and Eucommia ulmoides leaves contain chlorogenic acid,asperuloside,aucubin,rutin,geniposide,geniposidic acid and Quercetin,lignans and other medicinal active ingredients,which possesses the ability of liver protection,diuretic,cerebral hemorrhage prevention,anti-aging and blood pressure lowering etc.Meanwhile,it can cure a variety of other diseases as well.Chlorogenic acid,particularly,which is called "plant gold " internationally,is the main active ingredient in Eucommia ulmoides leaves.It bears the ability of scavenging free radicals,anti-virus,blood pressure lowering,tumor prevention and other biological activity.Moreover,the volume of chlorogenic acid in Eucommia ulmoides leaves is higher than that in Eucommia ulmoides.In addition,Eucommia ulmoides leaves also contains a variety of ingredients that Eucommia ulmoides do not.Eucommia are deciduous trees,the leaves of them are renewable and rich in resources.Therefore,Eucommia ulmoides leaves has great development and utilization value in the field of natural product research.With the continuous development of biochemical research,the use of enzyme engineering technology to extract the natural ingredients of Eucommia ulmoides had been approved by more and more researchers.Because the specificity of the enzyme,the protection of plants in the natural drug ingredients is particularly important.And the enzymatic conditions are rather moderate,which can protect the biological activity and efficacy of gutta-percha and natural medicine effectively.The raw materials do not need to be crushed,so there is no mechanical damage to the natural products,along with high dissolution of intracellular and extracellular natural products,achieving high efficiency extraction of main drug ingredients.This method was applied to the production practice based on the unique advantages of the enzyme,following work had been done:(1)Stepwise continuous enzymolysis,alcohol extraction and water extraction had been conducted to extract the main drug components in Eucommia ulmoides leaves.By comparing the three extraction methods,the optimum extraction methods were determined.(2)Establish HPLC(High Performance Lipuid Chromatopraphy)analysis method to gradient elution of active ingredients in the barks of Eucommia ulmoides oliv.The leaching conditions are optimized,and finally confirmed the best flow phase is 95% acetonitrile solution and 5% acetonitrile solution,and under the condition of gradient is:0-10min: 5% to 5% of B solution;10-35min: 5% to 40% of B solution;35-50min: 40% to 100%.Qualitative analysis of active ingredient that extract from enzyme solution,alcohol solution and water solution had been done.In addition,the enzyme solution is used to extract the asperuloside,aucubin,geniposide,geniposidic acid,quercetin and rutin.It can also extract substances that can’t be extracted by ethanol or water extraction.(3)At different pH,enzyme amount,temperature of protease,pectinase and cellulase respectively to extract the barks of Eucommia ulmoides oliv chlorogenic acid content.The experimental results show that protease optimum pH 3.4,adding amount of enzyme is 10%,the reaction temperature of 55 ℃,pectinase optimum pH 3.8,with enzyme amount was 5%,the reaction temperature is 50 ℃,cellulase optimum pH 4.8,adding amount of enzyme is 10%,the reaction temperature is 50 ℃.(4)Evaporation at different rotation speed,β-cyclodextrins amount,concentrated temperature on the impact of Eucommia ulmoides chlorogenic acid content in the powder,eucommia bark powder production process were optimized by response surface analysis method for making optimum process conditions of Eucommia ulmoides powder as 51r/min,β-cyclodextrins amount is 38.00 g,concentrated temperature is 61 ℃,under the condition of the chlorogenic acid content is 2.55μg/g.
